补肾活血颗粒对去势大鼠肾虚证的影响及机制研究

摘要
目的:探讨补肾活血颗粒对去势大鼠的肾虚症状的改善作用及作用机制,为临床防治骨质疏松症采用补肾治本的原则提供理论依据。方法:24只雌性大鼠单纯随机分为补肾活血组、模型组、假手术组,每组8只。补肾活血组与模型组麻醉后行双侧卵巢切除术,假手术组麻醉后行假手术处理,饲养3个月确认造模成功后各组分别给予补肾活血颗粒、生理盐水、生理盐水灌胃,90 d后进行3 d大鼠的一般情况观察及生理状态的量化检测,然后麻醉下处死大鼠,用电子天平称取垂体、肾、肾上腺、子宫的湿重,采用Elisa法进行ACTH、E2、T3、T4的血清学检测。结果:补肾活血组大鼠的体毛光滑略显稀疏、行动活泼、反应尚可、精神尚佳、食欲可、身体肥胖、略有嗜睡、眼睛明亮、眼角干燥、二便基本正常等表现;活动度、肛温及进食量均高于模型组,存在显著性差异(P<0.01),与假手术组无明显差异(P>0.05);血压与模型组及假手术组比较无明显差异(P>0.05);血清ACTH、E2、T3、T4的水平明显高于模型组,具有显著性差异(P<0.01),与假手术组无明显差异(P>0.05);垂体、肾上腺、肾的湿重均明显高于模型组,存在显著性差异(P<0.01),与假手术组比较无统计学意义(P>0.05),但子宫的湿重高于模型组,低于假手术组,均存在显著性差异(P<0.05)。结论:补肾活血颗粒能够明显的改善去势大鼠的肾虚症状,并且推测调节下丘脑-垂体-靶腺轴是补肾活血颗粒改善去势大鼠肾虚症状的作用机制。
        Objective: To observe kidney deficiency symptoms and the mechanism of ovariectomized rats treated with Bushen Huoxue Particles to provide a theoretical basis for clinical prevention and treatment of osteoporosis. Methods: 24 female SD rats were randomly divided into 3 groups: Bushen Huoxue Particles group,model group and sham group. Rats of Bushen Huoxue Particles group and model group were administrated with ovariectomy for establishing models of postmenopausal osteoporosis,while ovarian of sham group rats were exposed but were not removed. 3 groups were gavaged Bushen Huoxue Particles and saline for 3 months after confirming the model successfully. The general conditions were observed and physiological condition,estrogen( E2),adrenocorticotropic hormone( ACTH),three typical thyroid original acid( T3),four typical thyroid original acid( T4) and weight of kidney,adrenal gland,pituitary and uterus were detected in each group. Results: The general conditions of Bushen Huoxue Particles group was normal,for example,slightly sparse body hair with burnish,lively action,reaction agile,good spirit and appetite,obesity slight drowsiness,bright and dry eyes,normal defecation and urine. The activity,rectal temperature and eating were higher much more than that of the model group and there was statistical difference among these groups( P < 0. 01) but there was no significant difference compared with the sham group( P > 0. 05). The serum levels of E2,ACTH,T3 and T4 levels were significantly higher than those of the model group and there was statistical difference among these groups( P < 0. 01) but there was no significant difference compared with the sham group( P > 0. 05). The weights of pituitary,kidney and adrenal gland were significantly higher than those of the model group and there was statistical difference between these groups( P < 0. 01) but there was no significant difference compared with the sham group( P > 0. 05). The uterus was higher than that in the model group and lower than that in the sham group and there was difference among these groups( P < 0. 05). Conclusion: Bushen Huoxue Particles can significantly improve the kidney deficiency syndrome of the ovariectomized rats. We can speculate that the mechanism of Bushen Huoxue Particles improving the kidney empty symptoms of osteoporosis was the regulation of hypothalamic-pituitary-target gland axis.
