寒凝血瘀证裸小鼠模型的建立及评价

摘要
目的:探索建立寒凝血瘀证裸小鼠模型的方法,并对模型进行评价。方法:27只裸小鼠随机分为模型组和对照组,模型组(16只)采用盐酸肾上腺素皮下注射+冰水游泳制作寒凝血瘀证裸小鼠模型,对照组(11只)予等剂量生理盐水皮下注射。均持续14 d。观察两组裸小鼠的一般情况,记录体重、饮水量、摄食量,测定尾出血时间、凝血时间(玻管法)。结果:与对照组比较,模型组裸小鼠出现身体局部瘀点瘀斑、唇周黯红、舌色紫黯、嗜卧少动、行动迟缓、蜷卧喜扎堆、大便溏、小便清长及体重增长缓慢,饮水量、摄食量减少等表现;模型组裸小鼠尾出血时间、凝血时间(玻管法)均较对照组明显缩短。结论:盐酸肾上腺素皮下注射结合冰水游泳刺激可建立寒凝血瘀证裸小鼠模型,模型组裸小鼠符合寒凝血瘀证的特征性表现。
Objective:To establish and evaluate the nude mouse syndrome model of blood stasis due to cold accumulation.Methods:Twenty-sevent nude mice were randomly divided into model group and control group.The model group(16 nude mice) were given ice water bath and adrenaline subcutaneous injection to set up the nude mouse syndrome model of blood stasis due to cold accumulation,while the control group(11 nude mice) were given normal saline injection by the same dose.After 14-day continuous intervention,the general performance,weight,water intake,food intake,bleeding time of tail,blood coagulation time by glass tube were observed in all animals.Results:Model mice exhibited typical syndromes of blood stasis due to cold accumulation.They showed petechiae and ecchymosis somewhere,dark red color around the lip,dark purple of tongue color and lowered activities with more sleepiness,more love for collective,loose stool,clear abundant urine and slower weight gain and reduced water intake and food intake.Bleeding time of tail and blood coagulation time by glass tube of the model group were obviously shorter than that of the control group.Conclusion:Animals underwent intervention of ice water bath and adrenaline subcutaneous injection exhibited biological characteristics of syndrome of blood stasis due to cold accumulation,indicating that nude mice of blood stasis due to cold accumulation syndrome type were successfully made.
