七芍方对自发性高血压大鼠心室重构保护效应及其具体机制相关研究

摘要
目的 :观察七芍方对自发性高血压大鼠心室重构的保护效应,进一步探讨其具体机制。方法 :将40只13周龄雄性自发性高血压大鼠按照完全随机原则分为模型组、中药干预组、缬沙坦组,同时选取10只13周龄的Wistar大鼠设置空白对照组,其中中药干预组予七芍方灌胃,缬沙坦组予缬沙坦灌胃,模型组及空白对照组予相同体积的无菌用水灌胃,连续干预12周;予各组大鼠监测血压;采取免疫分析法对各组大鼠IL-1β、IL-10数值进行测定;采取Masson染色对心脏的形态学进行观察;采取Westernblot法测定心脏组织IL-1β、IL-10表达水平。结果 :与模型组大鼠对比,中药组大鼠血压下降显著(P<0.01);与模型组大鼠对比,中药组及缬沙坦组大鼠IL-1β数值下降,IL-10数值上升(P<0.01);与缬沙坦组大鼠对比,中药组大鼠IL-1β数值下降(P<0.05);与模型组大鼠对比,中药组及缬沙坦组心脏组织间质胶原原有增生显著减少;各组大鼠心脏组织光密度分析数值,中药组IL-1β数值显著低于缬沙坦组(P<0.05);Westernbiot法测定心脏组织蛋白表达水平,IL-1β半定量测定数值对比,中药组与缬沙坦组均显著较模型组大鼠下降(P<0.01);中药组显著较缬沙坦组数值下降(P<0.05);IL-10半定量测定数值对比,中药组及缬沙坦组均较模型组大鼠下降(P<0.05)。结论 :七芍方对自发性高血压大鼠心室重构具有显著的保护效应,具体机制可能与其能够使血浆及心脏组织间IL-1β含量下降、IL-10含量上升相关。
        Objective :To observe the protective effect of Qishao Decoction on ventricular remodeling in spontaneously hypertensive rats and further to explore its specific mechanism. Methods :Forty-five15-week-old male spontaneously hypertensive rats were divided into model group,Chinese medicine group(Qishao Decoction group),and valsartan group according to the principle of randomization. Twelve Wistar rats aged 15 weeks were selected to set up health. The rats in the traditional Chinese medicine group were given Qishao Decoction by gavage,the valsartan group was given valsartan by gavage,the model group and the healthy group were intragastrically administered with the same volume of water for 12 weeks,and blood pressure was monitored in each group. The number of IL-1β and IL-10 in each group was measured by immunoassay. The morphology of the heart was observed by Masson's staining. The levels of IL-1β and IL-10 in the heart tissue were measured by Western blot. Results :Compared with the model group,the blood pressure of the traditional Chinese medicine group decreased significantly(P<0.01). Compared with the model group rats,the IL-1β value of the Chinese medicine group and the valsartan group decreased,and the IL-10 value increased(P<0.01);Compared with the valsartan group,the IL-1β value of the rats in the Chinese medicine group was decreased(P<0.05). Compared with the model group rats,the original collagen hyperplasia of the cardiac tissue in the Chinese medicine group and the valsartan group significantly reduced;Densitometric analysis of heart tissue in each group,IL-1β in Chinese medicine group was significantly lower than that in valsartan group(P<0.05);Westernbiot method was used to measure protein expression in heart tissue,and IL-1β was semi-quantitatively measured,Chinese medicine group and valsartan group were significantly lower than the model group rats(P<0.01);Chinese medicine group was significantly lower than the valsartan group values(P<0.05);IL-10 semi-quantitative determination of numerical comparison,Chinese medicine group and valsartan group were lower than the model group rats(P<0.05). Conclusion :Qishao Decoction has significant protective effect on ventricular remodeling in spontaneously hypertensive rats. The specific mechanism may be related to the decrease of IL-1β content and the increase of IL-10 level in plasma and cardiac tissue.
