摘要
目的:研究电针疗法的早期介入对颅脑损伤大鼠认知功能的改善作用。方法:将30只雄性SD大鼠随机分为空白组、模型组和电针组各10只。采用改良的Feeney自由落体打击方法对模型组与电针组大鼠制造颅脑损伤(TBI)模型。术后给予抗炎抗感染治疗,另外对电针组大鼠进行电针治疗,穴位选取"人中""百会",患侧前肢"曲池""内关",患侧后肢"足三里""三阴交"等穴,连续治疗21 d。所有大鼠于术后1 d起进行改良神经功能缺损评分(mNSS)评估;16 d时开始水迷宫测试,计算各组大鼠水迷宫航行时间(潜伏期)、平台象限停留时间百分比;造模21 d后处死大鼠取出脑组织,采用苏木素-伊红染色观测并计算脑组织损伤体积百分比。结果:电针组较模型组在治疗后4~21 d m NSS得分差异有统计学意义(P<0.05);电针组较模型组在水迷宫测试2 d后就显著降低了水迷宫航行时间(潜伏期)的水平,差异有统计学意义(P<0.05),其平台象限停留时间百分比明显高于模型组,差异有统计学意义(P<0.01);21 d后电针组的脑损伤体积百分比明显小于模型组,差异有统计学意义(P<0.05)。结论:电针可明显修复TBI大鼠的神经功能,更好地改善其认知功能。
        Objective:To study the effect of early intervention of electroacupuncture on cognitive function in rats with craniocerebral injury. Methods:Thirty male Sprague-Dawley rats were randomly divided into blank group,model group and electroacupuncture group. The model of brain injury(TBI) was made in the model group and the electroacupuncture group using a modified Feeney free fall method. Anti-inflammatory and anti-infective treatment was given after operation. In addition,electroacupuncture treatment was performed on rats in the electroacupuncture group. The acupoints Shuigou(DU 26) and Baihui(DU 20) were selected,and also the acupoints of Quchi(LI 11),Neiguan(PC 6) on the forelimbs of the affected side,Zusanli(ST 36)and Sanyinjiao(SP 6) on the hind limb of affected side were selected. The acupuncture treatment lasted for21 days. All rats were evaluated for modified neurological severity score(mNSS) on the first day after surgery;water maze test started on the 16 th day,and the water maze navigation time(latency period) and the percentage of platform quadrant residence time of each group were calculated. On the 21 th day,the rats were sacrificed and brain tissues were sampled. The HE staining was used to observe and calculate the percentage of brain tissue damage volume. Results:Compared with the model group,on the m NSS score the electroacupuncture group was lower with statistical difference(P<0.05),on the 2 nd day after the water maze test on the water labyrinth sailing time(latency period) was reduced with statistical difference(P<0.05),on the plate quadrant residence time percentage was significantly higher with statistical difference(P<0.01),and the percentage of brain injury volume was significantly lower after 21 days of treatment with statistical difference(P<0.05). Conclusion:Electroacupuncture can significantly repair the neurological function of TBI rats and improve their cognitive function.
