摘要
目的 :观察小半夏汤对大鼠离体肠管运动的影响,探讨其对胃肠运动的作用及机制。方法 :将大鼠离体回肠收缩张力和振幅作为指标,分别选取乙酰胆碱(Acetylcholine chloride,ACh)、氯化钡(Barium chloride,BaCl_2)、阿托品(Atropine sulfate,Atr)、多巴胺(Dopamine,DA)、肾上腺素(Adrenaline,Adr)为工具药,利用生物信号采集处理系统,观察小半夏汤对各工具药所致大鼠回肠收缩张力和振幅的影响。结果 :小半夏汤可降低正常大鼠离体肠管的收缩张力和振幅,对大鼠离体回肠的自主运动具有抑制作用。小半夏汤高、中、低剂量均对ACh、BaCl_2引起的回肠张力升高、振幅变大有拮抗作用,使其张力和振幅降低。小半夏汤高、中剂量能使先用DA而张力下降的离体大鼠回肠进一步松弛,小半夏汤各剂量对Atr和Adr所致的回肠收缩抑制无明显影响。结论 :小半夏汤对肠管运动具有抑制作用。这种效应可能通过阻断M受体,激动多巴胺受体,或通过对平滑肌的直接作用而实现。
Objective :To observe the effect of Xiaobanxia Decoction(XBXD)on the movement of rat isolated ileum,and to investigate the effect and mechanism of XBXD on gastrointestinal motility. Methods :The effects of different concentrations of XBXD on ileum contraction in rats were observed by using the systolic tension and amplitude of isolated ileum in rats. Results :XBXD could reduce the contractile tension and amplitude of the isolated intestine in normal rats,and inhibit the autonomic movement of the isolated ileum in rats. All the three doses(20,10,4 g·L~(-1))of XBXD could suppress the contractility,significantly reduced the tone and amplitude of ileum contractions stimulated by acetylcholine chloride(ACh)and barium chloride(BaCl_2). All the three doses had additive effects on tone and amplitude of ileum contractions with DA. All the three doses had no effects on tone and amplitude of ileum contractions with adrenaline and atropine. Conclusion :XBXD has an inhibitory effect on the movement of intestinal tract. This effect may be achieved by blocking the M receptor,activating dopamine receptors,activating dopamine receptors,or by direct action on ileum smooth muscle. The mechanism of action of Xiaobanxia Decoction on gastrointestinal motility needs further study.
