葛根素治疗膝关节骨性关节炎模型兔的机制

摘要
目的:探讨葛根素治疗膝关节骨性关节炎模型兔的机制。方法:选取普通级成年新西兰白兔44只,按照随机数字表法分为正常对照组、模型对照组、生理盐水组和葛根素组,每组均为11只。除正常对照组外,其余3组采用改良Hulth模型法建造膝关节骨性关节炎动物模型。建模后正常对照组和模型对照组不做任何处理,生理盐水组在兔膝关节处注射生理盐水,葛根素组在兔膝关节注射葛根素,均连续注射10周。比较各组兔的关节腔病变程度,各组兔关节液中的白细胞介素-1β(interleukin-1β,IL-1β)、IL-10、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平,比较各组兔软骨组织中的基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)、MMP-13、基质金属蛋白酶组织抑制因子-1(tissue inhibitor of metalloproteinase-1,TIMP-1)水平。结果:葛根素组兔的关节腔病变程度高于正常对照组,低于模型对照组和生理盐水组(P<0.05);葛根素组关节液中IL-1β、MDA高于正常对照组,IL-1β、TNF-α、MDA水平低于模型对照组和生理盐水组,IL-10、SOD水平高于模型对照组和生理盐水组,差异均有统计学意义(P<0.05);葛根素组的软骨组织中MMP-1、MMP-13阳性细胞比例高于正常对照组,低于模型对照组和生理盐水组,TIMP-1阳性细胞比例低于正常对照组,高于模型对照组和生理盐水组,差异均有统计学意义(P<0.05)。结论:葛根素具有抗炎、抗氧化以及降低MMPs表达的作用,这可能是其治疗膝关节骨性关节炎的作用机制。
        Objective:To explore the mechanism of puerarin in the treatment of model rabbits with knee osteoarthritis.Methods:44 adult New Zealand white rabbits were randomly divided into normal control group,model control group,saline group and puerarin group according to the random number table method.Each group had 11 rabbits.Except the normal control group,the other three groups used the modified Hulth model method to construct animal models with knee osteoarthritis.Normal control group and model control group did not do any treatment after modeling.Normal saline group was injected with normal saline at the knee joint of rabbits,and puerarin group was injected with puerarin at the knee joint of rabbits.Both groups were injected continuously for 10 weeks.The degree of joint cavity lesions in each group was compared.The levels of interleukin-1β(IL-1β),IL-10,tumor necrosis factor-α(TNF-α),superoxide dismutase(SOD) and malondialdehyde(MDA) in the synovial fluid of rabbits in each group were compared.The levels of matrix metalloproteinase-1(MMP-1),MMP-13 and tissue inhibitor of metalloproteinase-1(TIMP-1) in cartilage tissues of rabbits in each group were compared.Results:The degree of joint cavity lesion in puerarin group was higher than that in normal control group,lower than that in model control group and saline group(P<0.05).The levels of IL-1β and MDA in joint fluid of puerarin group were higher than those of normal control group,while the levels of IL-1β,TNF-αand MDA were lower than those of model control group and saline group,and the levels of IL-10 and SOD were higher than those of model control group and saline group.The differences were statistically significant(P<0.05).The percentage of MMP-1 and MMP-13 positive cells in cartilage tissue of puerarin group was higher than that of normal control group,lower than that of model control group and saline group,and the percentage of TIMP-1 positive cells was lower than that of normal control group,higher than that of model control group and saline group.The differences were statistically significant(P<0.05).Conclusion:Puerarin has anti-inflammatory,anti-oxidative and anti-MMPs expression,which may be the mechanism of Puerarin in the treatment of knee osteoarthritis.
