摘要
目的:探讨敦煌固本方对脾虚型肠易激综合征(D-IBS)大鼠结肠黏膜肥大细胞、类胰蛋白酶以及5-羟色胺(5-HT)的影响。方法:采用束缚+灌番泻叶造D-IBS大鼠模型,分为对照组、模型组、阳性药物组、敦煌固本方低、高剂量组,ELISA实验检测各组大鼠血清中5-HT含量变化;免疫组化实验检测各组大鼠结肠组织中类胰蛋白酶表达变化;甲苯胺蓝染色观察各组大鼠结肠组织中肥大细胞数量变化。结果:与模型组比较,敦煌固本方高剂量组血清中5-HT含量显著降低(P<0.01),类胰蛋白酶含量、肥大细胞数量在敦煌固本方高剂量组结肠组织中显著降低(P<0.01)。结论:敦煌固本方可通过减少D-IBS大鼠结肠组织中肥大细胞数量,进而使结肠组织中类胰蛋白酶含量降低,并使D-IBS大鼠血清中5-HT含量降低,从而达到对D-IBS的治疗作用。
Objective: To investigate the effect of Dunhuang Guben Formula on colonic mucosal mast cells, tryptase and serotonin(5-hydroxytryptamine) in rats with spleen deficiency irritable bowel syndrome(D-IBS). Methods: The rat model of D-IBS was established by restraint+senna. The changes of serum 5-HT content in the control group, model group, positive drug group, Dunhuang Guben Formula low-dose group and high-dose group were detected by ELISA. Immunohistochemistry was used to detect the expression of tryptase in colon tissue of each group. Toluidine blue staining was used to observe the changes of mast cells in colon tissue of each group. Results: Compared with the model group, the content of 5-HT in the group of Dunhuang Guben Formula high dose group was significantly decreased(P<0.01), and the content of tryptase was significantly decreased in the colon tissue of Dunhuang Guben Formula high dose group(P<0.01). The number of mast cells was significantly decreased in the colon tissue of Dunhuang Guben Formula high-dose group(P<0.01). Conclusion: Dunhuang Guben Formula can reduce the content of tryptase in colon tissue by reducing the number of mast cells in colon tissue of D-IBS rats, and reduce the content of 5-HT in serum of D-IBS rats, which achieves the therapeutic effect of D-IBS.
