摘要
目的:观察疮灵液对鼠永生化细胞(ATCC~?CRL-2770TM)黑素细胞黑素生成的作用。方法:应用MTT法测鼠永生化黑素细胞增殖、多巴氧化法测鼠永生化黑素细胞内酪氨酸酶活性、NaOH裂解法测鼠永生化黑素细胞黑素含量。结果:疮灵液在100~1 600μg/mL剂量,对小鼠永生化黑素细胞增殖具有增进作用,400μg/mL剂量促进作用最明显(P<0.05)。疮灵液为800μg/mL~3 200μg/mL剂量,可显著激活鼠永生化黑素细胞酪氨酸酶活性(P<0.01)。疮灵液于100μg/mL~3 200μg/mL剂量,对鼠永生化黑素细胞黑素合成均有增进作用(P<0.05)。结论:疮灵液对鼠永生化黑素细胞黑素的合成具有显著增进作用,并且在治疗诸如白癜风的色素疾病中具有潜在的应用。
Objective: To observe the effect of Chuangling Liquid on the melanogenesis of melanocytes in rat immortalized cells(ATCC~? CRL-2770 TM). Methods: Melanocyte proliferation was determined by MTT assay, tyrosinase activity of melanocyte was detected by dopa oxidation method, and melanin content was measured by NaOH lysis method. Results: When the concentration of Chuangling Liquid was at 100~1 600 μg/mL, it promoted the proliferation of melanocytes, and the promotion effect was most obvious at 400 μg/mL(P<0.05). Tyrosinase activity was significantly activated when the concentration of Chuangling Liquid was at 400 μg/mL(P<0.01). When the concentration of Chuangling Liquid was at 100~3 200 μg/mL, it promoted melanin synthesis(P<0.05). Conclusion: Chuangling Liquid has a promoting effect on melanin synthesis of melanocyte, which has a promising effect on leukoderma of hypopigmentation disease.
