摘要
目的:以药效学为指标对苓仙降酸复方的不同工艺提取物进行比较研究,筛选出最佳提取工艺。方法:通过对Wistar大鼠灌胃腺嘌呤和氧嗪酸钾建立高尿酸血症模型,将40只建模成功的大鼠随机分为模型组,苓仙降酸复方水提组(10g生药/kg)、水提醇沉组(10g生药/kg)、醇提组(10g生药/kg),阳性对照别嘌醇组(20mg/kg),另设空白对照组。每组均灌胃给药,qd,共21d。于第7、14、21天由大鼠眼眶取血测定尿酸(UA)、尿素氮(BUN)、血肌酐(CR)水平,第21天取血后处死大鼠,取其脏器,测定肝脏中腺苷脱氨酶(ADA)水平和称量肾脏重量。结果:与模型组比较,苓仙降酸复方水提组大鼠的UA、BUN、CR、肾脏重量和ADA水平均显著降低(P<0.05,P<0.01),而水提醇沉组和醇提组的各项指标部分有显著差异,部分仅有降低趋势。结论:苓仙降酸复方水提物降UA及缓解肾脏病理损伤的作用优于水提醇沉物和醇提物,其最佳提取工艺为水提取。
        Objective:To study the extracts from Lingxianjiangsuan compound by different extraction methods with pharmacodynamics as an index,so as to screen out the optimal extraction method.Methods:The Wistar rats were gavaged with adenine and potassium oxonate to establish hyperuricemia models,and then upon successful establishment of the model,40 rats were randomly divided into the model group,the Lingxianjiangsuan compound water extraction group,the water extraction and alcohol precipitation group,the alcohol extraction group(10 g crude drugs/kg),the positive controlled allopurinol treatment group(20 mg/kg)and the blank control group.The drugs were administered by gavage once daily for 21 d.The levels of uric acid(UA),urea nitrogen(BUN),serum creatinine(CR)were determined on days 7,14 and 21,respectively.On day 21,the rats were excuted,after blood samples were taken,and viscera were obtained.Then the level of adenosine deaminase(ADA)in liver and kidney weight were measured.Results:Compared with those of the model group,the levels of UA,BUN and CR,kidney weight,level of ADA in liver of the water extraction group were significantly decreased(P<0.05,P<0.01),while some indicators in the water extraction and alcohol precipitation group and the alcohol extraction group were significantly difference,and some indicators showed a decreasing trend.Conclusion:The water extract of Lingxianjiangsuan compound has better effects on the reduction of UA and alleviation of pathological damage to kidney,as compared with the water extraction and alcohol precipitation and alcohol extraction,and the optimal extraction method of Lingxianjiangsuan compound is water extraction.
