输尿管不全梗阻时段平滑肌肌球蛋白表达的研究

摘要
目的探讨输尿管不全梗阻时间与平滑肌肌球蛋白表达量的关系。方法 120只SD大鼠随机分为正常组30只,模型组与实验组各45只;模型组麻醉后切开腹部,游离右侧输尿管下段约2. 0 cm,分离出右侧腰大肌隧道,不做包埋,缝合切口;实验组在模型组基础上实施右侧输尿管下段腰大肌隧道包埋;术后1、2、3周各组随机处死部分大鼠,观察不同时段输尿管形态结构变化并检测平滑肌肌球蛋白表达。结果正常组与模型组右侧输尿管直径比较差异无统计学意义(P>0. 05),其与实验组比较差异有统计学(P <0. 01);实验组梗阻近端输尿管随时间延长增粗,第3周最为显著(P <0. 01),平滑肌肌球蛋白表达也逐渐增加,2周时明显,第3周却显著降低,低于正常组与模型组(P <0. 01)。结论输尿管不全梗阻超过2周,平滑肌肌球蛋白表达显著降低,管壁不可逆性改变较为明显。
Objective To investigate the relationship between the time of ureteral obstruction and the expression of smooth muscle myosin. Methods A total of 120 Sprague-Dawley rats was randomly divided into normal group( n = 30),model group and experimental group( n = 45). The model group was incised in the abdomen after anesthesia and the right lower ureter was dissociated about 2. 0 cm and the right psoas muscle tunnel was separated. The lower part of the pneumothorax embedded in the right ureter was performed in experimental group on the basis of model group. One,2 and 3 weeks later,rats were sacrificed and the ureteral morphological changes and smooth muscle myosin expression were detected,respectively. Results There was significant difference in the diameter of the right ureter between the model and the experimental groups with the proximal ureter obstruction thickened in the experimental group 3 weeks after the model established. Compared with the model group,smooth muscle myosin expression in the experimental group was increased in 2 weeks after ureteral obstruction and decreased at 3 weeks after ureteral obstruction. Conclusion More than 2 weeks after ureteral obstruction,smooth muscle myosin expression was significantly reduced and the irreversible wall changes were more obvious.
