摘要
目的:分析薤白多糖提取、分离提纯及与DNA作用。方法:根据薤白多糖酶法提取工艺、对薤白多糖薤进行分离提纯和鉴定。研究期间利用常规方法分离小鼠股骨BMSCs,将BMSC分为空白组、对照组、实验组(剂量100μL、剂量为12.5、25、50、100μg/ml)薤白多糖,分别采用不同方法处理。结果:AMP40-1、AMP40-2、AMP60-1、AMP80-1均未见吸收峰,提示为单纯的多糖组份。空白组、对照组、不同浓度的薤白多糖溶液处理的实验组的畸变率、焦点簇数目差异都具有统计学意义(P<0.05)。结论:薤白多糖提取、分离提纯质量尚可;薤白多糖可减轻放射引起的细胞DNA损伤,发挥保护作用且呈剂量依赖。
        Objective: to analyze the extraction, separation and purification of polysaccharide from Allium macrostemon Bunge and its interaction with DNA. Methods: according to the enzymatic extraction process of Allium macrostemon polysaccharide, the polysaccharide of Allium macrostemon Bunge was purified and identified. During the study period, BMSC was isolated from mouse femur BMSCs, by routine method. BMSC was divided into blank group and control group. The experimental group was treated with different methods(100 μl, 12.5 μg/ml). Results: no absorption peak was found in AMP40-1,AMP40-2,AMP60-1,AMP80-1, indicating that it was a simple polysaccharide component. Blank group In the control group, there were significant differences in the aberration rate and the number of focal clusters in the experimental group treated with different concentrations of Allium macrostemon polysaccharide solution(P < 0.05). Conclusion: the polysaccharides of Allium macrostemon Bunge can be extracted and purified, and the polysaccharide of Allium macrostemon can attenuate the DNA damage induced by radiation and play a protective role in a dose-dependent manner.
