摘要
[目的]采用响应面法优化超声提取黄荆叶总黄酮的工艺条件及月周期规律研究,为其开发利用提供技术参考。[方法]以9月份的黄荆叶粗粉为原料进行优化提取工艺研究,乙醇为提取溶剂,在单因素试验结果的基础上,以黄荆叶总黄酮提取率为响应值,应用响应面法考察提取次数、料液比、乙醇浓度和超声功率对黄荆叶总黄酮提取率的影响,根据Box-Behnken中心组合试验设计原理,设计4因素3水平响应面试验,建立回归模型,进行响应面分析,并根据最优提取工艺提取4—11月黄荆叶的总黄酮,确定总黄酮含量的变化规律。[结果]9月份黄荆叶总黄酮超声提取最佳工艺条件为:提取次数为3次,料液比为1.00∶50.25,乙醇浓度为50.59%,超声功率比为41.94%,总黄酮提取得率可达10.48%;4—11月份中8月份的黄荆叶总黄酮含量最高,可达11.22%。[结论]响应面法优化黄荆叶总黄酮的提取条件切实可行,得到的回归方程与试验结果拟合性好,方法稳定可靠,可为工业大生产提取黄荆叶总黄酮提供一定的技术参考。
        [Objective]The response surface methodology was used to optimize the extraction process and monthly cycle law of total flavonoids from Vitex negundo leaves, and provided technical reference for its development and utilization. [Method]Based on the optimization of the extraction process of the yellow wattle leaf in September, ethanol was used as the extraction solvent. Based on the single factor test results, the extraction rate of total flavonoids from Vitex negundo was used as the response value, and the response times were used to investigate the extraction times and solid-liquid ratio. The effects of ethanol concentration and ultrasonic power on the extraction rate of total flavonoids from Vitex negundo leaves were studied. According to the design principle of Box-Behnken center combination test, a 4-factors 3-levels response surface test was designed, a regression model was established, response surface analysis was performed, and according to the optimal extraction process. The total flavonoids of Vitex negundo leaves were extracted from April to November, and the highest month was determined. [Result]The optimum conditions for ultrasonic extraction of total flavonoids from Vitex negundo in September were as follows: extraction times were 3 times, solid-liquid ratio was 1.00∶50.25, ethanol concentration was 50.59%, ultrasonic power was 41.94%, and total flavonoid extraction yield was 10.48%;In the middle of April-November, the total flavonoids content of Vitex negundo leaves in August was the highest, reaching 11.22%.[Conclusion]It is feasible to optimize the extraction conditions of total flavonoids from Vitex negundo by response surface methodology.The regression equation obtained was well fitted with the experimental results, and the methodology was stable and reliable. It could provide a technical reference for the industrial production of total flavonoids from Vitex negundo.
