摘要
目的:探讨穴位埋线法对围绝经期大鼠脂质代谢的影响及其机制。方法:利用双侧卵巢切除(OVX)的方法制备围绝经期大鼠模型,SD大鼠随机分为假手术组、模型组、雌激素组和埋线组,每组10只。比较各组大鼠体质量变化、子宫系数、大鼠血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)和高密度脂蛋白(HDL)及肝脏组织中腺苷酸活化蛋白激酶(AMPKα)、磷酸化腺苷酸活化蛋白激酶(p-AMPKα)、乙酰辅酶A羧化酶(ACC)及磷酸化乙酰辅酶A羧化酶(p-ACC)蛋白表达。结果:与假手术组相比,模型组大鼠体质量明显升高、子宫系数却明显降低、血清中TC、TG、LDL含量均升高,而HDL含量下降、肝脏组织中p-AMPKα、p-ACC表达均有所下降,具有显著性差异(P<0.01)。经穴位埋线治疗后,大鼠体质量明显降低、子宫系数升高、血清中TC、TG、LDL含量均降低,而HDL含升高、肝脏组织中p-AMPKα、p-ACC表达均有所升高,具有显著性差异(P<0.05,P<0.01)。结论:穴位埋线法能够改善围绝经期大鼠脂质代谢紊乱,其可能的机制是通过激活AMPK信号通路,促进AMPKα、ACC磷酸化实现的。
        Objective: To explore the effect of acupoint catgut-embedding therapy on lipid metabolism and its possible mechanism in the perimenopendal rats. Methods: SD rats were randomly divided into the sham group, the model group, the estrogen group and the acupoint catgut-embedding therapy group, 10 mice in each group. In this study, the perimenopausal model in rats was established by Ovariectomy. Compared the differences in rats weight, uterus coefficient, total cholesterol(TC), triglyceride(TG), low density lipoprotein(LDL) and high-density lipoprotein(HDL) levels and the expressions of AMPK α, p-AMPK α, ACC and p-ACC were examined by Western Blot. Results: Compared with the sham group, model group's rats weight increased significantly, uterus coefficient was lower significantly, and TC, TG and LDL levels were increased, and HDL levels were decreased. The expressions of p-AMPK α and p-ACC proteins were statistically decreasedcompared with that of the sham group(P<0.01). After acupoint catgut-embedding therapy, the weight of ratssignificantly reduced. The uterus coefficient increased. TC, TG and LDL levels were decreasedand HDL levelsincreased. The expressions of p-AMPK α and p-ACC protein were increasedwith significant difference(P<0.05, P<0.01). Conclusion: The acupoint catgut-embedding therapy improved the lipid metabolism disorder in the perimenopaual ratsand its possible mechanism is to activate AMPK signaling pathway to promote AMPK α and ACC phosphorylation.
