β-榄香烯对STZ致糖尿病性视网膜病变大鼠增殖期SIRT1和VEGF表达的影响

摘要
目的研究β-榄香烯对链脲佐菌素(STZ)致糖尿病性视网膜病变大鼠增殖期沉默信息调节因子(SIRT1)和血管内皮生长因子(VEGF)表达的影响。方法选取80只Wistar大鼠作为研究对象,将其分为正常对照组、DR模型组、无菌柠檬酸组以及β-榄香烯组各20只。其中DR模型组、无菌柠檬酸组以及β-榄香烯组大鼠均经适应性喂养后通过腹腔一次性注射STZ制作糖尿病视网膜病变增殖期模型。无菌柠檬酸组大鼠造模成功后予以无菌柠檬酸腹腔注射干预,β-榄香烯组大鼠造模成功后则予以玻璃体腔内注射β-榄香烯干预。分别比较各组大鼠视网膜中SIRT1/β-actin相对灰度值、视网膜血管VEGF表达。结果 DR模型组、无菌柠檬酸组、β-榄香烯组以及正常对照组大鼠视网膜中SIRT1/β-actin相对灰度值分别为(1.218±0.033)、(1.305±0.052)、(1.411±0.088)、(1.502±0.103),呈逐渐升高趋势,且经单因素方差分析可得:组间比较差异均有统计学意义(P<0.05)。DR模型组、无菌柠檬酸组、β-榄香烯组以及正常对照组大鼠视网膜血管VEGF表达水平分别为(89.084±1.881)pg/m L、(57.920±0.910)pg/m L、(50.144±0.721)pg/m L、(44.382±0.661)pg/m L,呈逐渐下降趋势,且经单因素方差分析可得:组间比较差异均有统计学意义(P<0.05)。结论β-榄香烯对STZ致糖尿病性视网膜病变大鼠增殖期SIRT1表达具有一定的促进作用,而对VEGF表达具有明显的抑制作用。
        Objective To study the effect of β-elemene on the expression of SIRT1 and VEGF in diabetic retinopathy rats induced by streptozotocin(STZ). Methods 80 Wistar rats were selected as study subjects and divided into normal control group, DR model group, sterile citric acid group and β-elemene group, with 20 cases in each group. Among them, DR model group, sterile citric acid group and β-elemene group were all fed with STZ to produce proliferative phase model of diabetic retinopathy. Sterile citric acid group was injected intraperitoneally into the rats after successful modeling, and β-elemene group was injected with β-elemene intravitreally after successful modeling. The relative gray levels of SIRT1/β-actin in the retina and the expression of VEGF in the retinal vessels were compared in each group.Results The relative gray values of SIRT1/β-actin in retinas of DR model group, sterile citric acid group, β-elemene group and normal control group were(1.218±0.033),(1.305±0.052), 1.411±0.088) and(1.502±0.103) respectively, showing a gradually increasing trend. There was a statistically significant difference between groups by one-way analysis of variance(P<0.05). The expression levels of VEGF in the retinal blood vessels of DR model group, sterile citric acid group,β-elemene group and normal control group were(89.084 ±1.881) pg/m L,(57.920 ±0.910)pg/m L,(50.144 ±0.721)pg/m L and(44.382±0.661)pg/m L respectively, showing a gradually decreasing trend. And there was a statistically significant difference between groups by one-way analysis of variance(P<0.05). Conclusion β-elemene has a certain role in promoting the expression of SIRT1 in rats with STZ-induced diabetic retinopathy and has a significant inhibitory effect on the expression of VEGF.
