摘要
目的探讨七氟醚对大鼠海马组织N-甲基-D-天冬氨酸-2B受体(NR2B)、半胱氨酸蛋白酶3(Caspase-3)的影响。方法选取7d龄SD大鼠60只分为对照组和麻醉组,每组再分为3个时间点,即麻醉前、麻醉后24h及麻醉后7周,每组10只。麻醉组给予2.5%七氟醚麻醉4h,对照组仅给予空氧混合气。采用Western blot检测各组大鼠海马组织NR2B、Caspase-3蛋白表达,Morris水迷宫检测麻醉后7周大鼠的空间学习记忆能力。结果麻醉后24h,麻醉组大鼠海马组织NR2B、Caspase-3蛋白表达明显高于对照组及麻醉前(P<0.05);麻醉前及麻醉后7周,麻醉组和对照组大鼠海马组织NR2B、Caspase-3蛋白表达比较差异无统计学意义(P>0.05)。麻醉组大鼠第1、3天游泳潜伏期分别为(70.23±8.10)s和(41.03±7.39)s,总路程分别为(18.20±2.02)cm和(11.03±1.98)cm,明显高于对照组(P<0.05);麻醉组和对照组大鼠游泳速度比较差异无统计学意义(P>0.05)。结论七氟醚麻醉可即刻造成幼鼠海马组织NR2B、Caspase-3蛋白表达升高,同时其成年后的学习记忆能力有一定损伤,但成年后海马组织NR2B、Caspase-3蛋白表达无明显异常。
Objective To investigate the effect of sevoflurane on NR2B and Caspase-3 in rat hippocampal tissue.Methods Sixty SD rats of 7 dold were selected and divided into the control group and the anesthesia group,then each group was re-divided into 3 subgroups according to 3 timepoints,before anesthesia,24 h and 7 weeks after anesthesia,with 10 cases in each group.The anesthesia group was given 2.5% sevoflurane for 4 h,and the control group was given air oxygen mixture only.The expression of NR2B and Caspase-3 protein in hippocampal tissue of each group was detected by Western blot,and the rat spatial learning and memory ability in 7 weeks after anesthesia were detected by Morris water maze.Results The expression levels of NR2B protein and Caspase-3 protein in the hippocampus tissue at 24 hafter anesthesia in the anesthesia group were were significantly higher than those in the control group and before anesthesia(P<0.05).The expression levels of NR2B protein and Caspase-3 protein before anesthesia and at 7 weeks after anesthesia had no statistically significant difference between the anesthesia group and the control group(P>0.05).The swimming latency periods at 1,3 din the anesthesia group were(70.23±8.10)s and(41.03±7.39)s,and the total journeys were(18.20±2.02)cm and(11.03±1.98)cm respectively,which were significantly higher than those in the control group(P<0.05).There was no statistically significant difference in swimming speed between the anaesthesia group and the control group(P>0.05).Conclusion Sevoflurane anesthesia can instantly cause the increase of NR2B and Caspase-3 protein in the hippocampal tissue of young rats,meanwhile the learning and memory ability after adult has a certain damage,however,there is no obvious abnormality in hippocampus tissue NR2B and Caspase-3 protein expression after adult.
