摘要
目的观察脐疗对寒凝血瘀型痛经大鼠神阙穴区Toll样受体2(Toll-like Receptors,TLR2)表达的影响,初步探索脐疗效应与神阙穴区的关系,阐释部分启动机制。方法将50只雌性大鼠随机分为正常对照组、模型组、药物贴脐组、隔药灸脐组、艾条灸脐组,每组10只。除正常对照组外,其余各组均构建寒凝血瘀痛经模型,给予相应的处理及治疗。通过免疫荧光法、蛋白质免疫印迹法、实时荧光定量PCR技术检测各组大鼠神阙穴区TLR2的表达。结果药物贴脐组、隔药灸脐组及艾条灸脐组TLR2蛋白表达量较模型组均升高,艾条灸脐组TLR2蛋白表达量高于隔药灸脐组、药物贴脐组(P<0.01);药物贴脐组、隔药灸脐组及艾条灸脐组TLR2mRNA表达较模型组均明显升高(P<0.01),艾条灸脐组高于隔药灸脐组和药物贴脐组(P<0.01)。结论脐疗通过上调寒凝血瘀型痛经大鼠神阙穴区TLR2蛋白及mRNA表达以激活腧穴功能,是诱导神阙信息启动的部分机制。
        Objective To observe the effect of navel therapy on the expression of(Toll-like Receptors 2)TLR2 in Shenque of coagulated cold and blood stasis type dysmenorrhea model rats, to initially explore the relationship between the umbilical effect and Shenque, and explain part of the initiation mechanism. Methods Fifty female rats were randomly divided into five groups: normal control group, model group, drug sticking navel group,Herb-partitioned moxibustion on navel group, moxa moxibustion navel group, 10 in each group. In addition to normal control group, coagulated Cold and Blood Stasis dysmenorrhea model was constructed. We gave esch group the corresponding treatment. The expression of TLR2 in Shenque was detected by immunofluorescence, Western blotting and Quantitative Real-time PCR. Results The expression of TLR2 in the navel treatment groups were higher than that in model group. The expression of TLR2 in moxa moxibustion navel group was higher than that in drug sticking navel group and Herb-partitioned moxibustion on navel group(P<0.01);the expression of TLR2 mRNA in the navel treatment groups were higher than that in model group(P<0.01).The expression of TLR2 mRNA in moxa moxibustion navel group was higher than that in drug sticking navel group and Herb-partitioned moxibustion on navel group(P<0.01). Conclusion Navel therapy activates the acupoint function by up-regulating the expression of TLR2 and TLR2 mRNA in the Shenque of coagulated Cold and Blood Stasis type dysmenorrhoea rat, is part of the mechanism for inducing the initiation of Shenque information.
