“以俞调枢”法对胃肠动力障碍大鼠SCF、c-kit蛋白及mRNA表达的影响

摘要
目的:观察"以俞调枢"法对胃肠动力障碍模型大鼠干细胞因子(SCF)及其受体(c-kit)蛋白及mRNA表达的影响,探索该疗法改善胃肠动力的作用机制。方法:选择SD大鼠120只,随机分为空白对照组、空白治疗组、模型对照组和模型治疗组各30只。空白对照组、模型对照组每日固定22min,不实施背俞指针治疗;空白治疗组、模型治疗组每日固定22min,并实施背俞指针治疗。疗程均为14d,结束后观察并比较4组大鼠胃内残留率、小肠推进比、SCF蛋白、c-kit蛋白、SCF mRNA、c-kit mRNA的变化。结果:模型对照组与空白对照组比较,其胃内残留率显著升高,小肠推进比率显著降低(P<0.01);模型治疗组与模型对照组比较,胃内残留率减少,小肠推进比增加(P<0.01)。与空白对照组比较,模型对照组消化道平滑肌组织中SCF、c-kit蛋白及mRNA减少(P<0.01);与模型对照组比较,模型治疗组上述指标增加(P<0.05,P<0.01)。结论:"以俞调枢"法具有较好的改善胃肠动力的疗效,其机制可能与其通过调节SCF、c-kit蛋白及mRNA的表达有关。
        Objective: To observe the effects of ‘regulating the pivot through the shu points' on the expression of SCF,c-kit protein and mRNA in rats with gastrointestinal motility disorder, and to explore the mechanism of this therapy to improve gastrointestinal motility. Methods: One hundred and twenty SD rats were randomly divided into blank control group, blank treatment group, model control group and model treatment group. The blank control group and the model control group were fixed for 22 minutes every day, and the treatment of the back-shu pointer was not performed. The blank treatment group and the model treatment group were fixed for 22 minutes every day, and the treatment of the back-shu pointer was performed. The course of treatment was 14 days. After the experiment, the changes of gastric residual rate, small intestinal propulsion ratio, SCF protein,c-kit protein, SCF mRNA and c-kit mRNA were observed and compared. Results: Compared with the blank control group, the gastric residual rate of model treatment group was increased, and the small intestinal propulsion rate of model treatment group was significantly decreased(P<0.01). Compared with the model control group, the gastric residual rate of model treatment group was decreased, and the small intestinal propulsion rate of model treatment group was significantly increased(P<0.01). Compared with the blank control group, the protein and mRNA expressions of SCF, c-kit in model control group were significantly decreased(P<0.01). Compared with the model control group, the protein and mRNA expressions of SCF, c-kit in model treatment group were significantly increased(P<0.01). Conclusion: The ‘regulating the pivot through the shu points' method has a better effect on improving gastrointestinal motility, and its mechanism may be related to the regulation of SCF, c-kit protein and mRNA expression.
