摘要
目的探讨子宫内膜癌组织中长链非编码RNA H19(LncRNA H19)、微小RNA-29b(miR-29b)、微小RNA-124(miR-124)表达水平,以及其与子宫内膜癌临床特征及预后的关系。方法选取2012年9月至2015年9月湖北省荆州市第三人民医院收治的90例行子宫切除术的子宫内膜癌患者为受试对象,术中留取子宫内膜癌组织和相应癌旁组织,另选取同期因子宫肌瘤切除子宫的85例正常子宫内膜组织(经病理检查证实)。采用实时荧光定量聚合酶链反应(qRT-PCR)检测组织中LncRNA H19、miR-29b、miR-124表达情况,分析它们的表达水平与子宫内膜癌患者临床特征的关系,并观察3年总生存率(OS)。结果子宫内膜癌组织LncRNA H19相对表达量显著高于癌旁组织和正常子宫内膜组织(P<0.05),miR-29b、miR-124相对表达量明显低于癌旁组织和正常子宫内膜组织(P<0.05)。LncRNA H19、miR-29b、miR-124表达水平与子宫内膜癌患者国际妇产科联合会(FIGO)分期、病理分级、子宫肌层浸润深度及盆腔淋巴结转移有关(P<0.05)。子宫内膜癌患者LncRNA H19表达水平与miR-29b、miR-124均呈负相关(P<0.05)。LncRNA H19低表达患者3年OS显著高于高表达患者(P<0.05),miR-29b、miR-124高表达患者3年OS明显高于低表达患者(P<0.05)。FIGO分期、子宫肌层浸润深度,以及LncRNA H19、miR-29b、miR-124表达水平是影响子宫内膜癌患者预后的独立危险因素。结论子宫内膜癌组织中LncRNA H19高表达,miR-29b、miR-124低表达。
        Objective To investigate the expressions of long non-coding RNA H19(LncRNA H19),micro RNA 29b(miR-29b)and micro RNA 124(miR-124)in endometrial carcinoma tissues and their relationship with clinical features and prognosis of endometrial cancer.Methods Ninety patients with endometrial cancer treated by hysterectomy in the Jingzhou Municipal Third People′s Hospital from September 2012 to September 2015 were selected as the subjects.The endometrial cancer tissues and corresponding paracancerous tissues were preserved during operation,other contemporaneous 85 cases of normal endometrium tissue due to hysteromyoma resection(confirmed by pathological examination)were selected.The qRT-PCR method was adopted to detect the expressions of LncRNA H19,miR-29b and miR-124 in tissues,their expressions in different tissues and their relationships with the clinical characteristics in the patients with endometrial cancer were analyzed,moreover the 3-year overall survival rate(OS)was observed.Results The relative expression level of LncRNA H19 in endometrial carcinoma tissues was significantly higher than that in paracancerous tissues and normal endometrial tissues(P<0.05),the relative expression levels of miR-29 band miR-124 were significantly lower than those in paracancerous tissues and normal endometrium tissues(P<0.05).The expression levels of LncRNA H19,miR-29 band miR-124 were related with FIGO stage,pathological grade,depth of myometrial invasion and pelvic lymph node metastasis(P<0.05).The expression level of LncRNA H19 was negatively correlated with miR-29b and miR-124 expressions in the patients with endometrial carcinoma(P<0.05).The 3-year OS in the patients with low expression of LncRNA H19 was significantly higher than that in the patients with high expression(P<0.05),the 3-year OS in the patients with high expressions of miR-29b and miR-124 was significantly higher than those in the patients with low expressions(P<0.05).FIGO staging,myometrial invasion depth,LncRNA H19,miR-29b and miR-124 expressions were independent risk factors affecting the prognosis in the patients with endometrial cancer.Conclusion LncRNA H19 in endometrial carcinoma tissue is highly expressed,while miR-29b and miR-124 are lowly expressed.
