基于单味药-配伍的合欢花与酸枣仁水提取物对抑郁性失眠动物模型的影响

摘要
目的对酸枣仁、合欢花水提取物单味药和配伍应用进行抗抑郁性失眠研究,探讨其配伍应用及作用机制。方法采用抑郁性失眠动物模型,观察酸枣仁、合欢花水提取物单味药和配伍应用对行为学、海马组织内单胺神经递质和氨基酸神经递质含量及比值的影响。结果酸枣仁-合欢花提取物高、低剂量组及其单味药提取物高、低剂量组与模型组比较,旷场实验水平得分与垂直得分都明显提高(P <0.01,P <0.05)或有升高趋势,海马组织中单胺神经递质含量也明显增高(P <0.001,P <0.01)或有增高趋势,氨基酸神经递质Glu含量及Glu/GABA明显降低(P <0.001,P <0.01,P <0.05)或有降低趋势,GABA含量明显增高(P <0.01,P <0.05)或有增高趋势。结论酸枣仁、合欢花提取物单独或配伍应用,都能发挥改善抑郁性失眠的功效,并以配伍应用更佳。
        Objective To study the use of water extract of Flos Albiziae and Semen Ziziphi Spinosae on anti-sleep deprivation depression based on single herb and compatibility, and to probe into the effect and mechanism of action of compatibility. Methods We adopted sleep deprivation depressionanimal model, to observe the effecton behavio ristics,monamineneurotransmitter content and amino acid neurotransmitters content and their ratio in hippocampus of the animals with the use of water extract of Flos Albiziae and Semen Ziziphi Spinosae based on single herb and compatibility. Results Compared with the model group, horizontalscore and verticalscore in open field test were obviously enhanced of the rats in Flos Albiziae and Semen Ziziphi Spinosae compatibility high-dose and low-dose groups and Flos Albiziae and Semen Ziziphi Spinosae single herb high-dose and low-dose groups(P < 0.01, P < 0.05)or had rising trend, and their monamine neurotransmitter content in hippocampus were obviously rised(P < 0.001,P < 0.01) or had rising trend, and their amino acid neurotransmitters content and their ratio in hippocampus were obviously reduced(P < 0.001, P < 0.01, P < 0.05) or had lower trend. The content of GABA increased significantly(P < 0.01, P < 0.05). Conclusion Whether compatibility use or single herb use of water extract of Flos Albiziae and Semen Ziziphi Spinosae, they all could develope anti-sleep deprivation depression effect. Compatibility use was better than single herb use.
