星点设计-效应面法优化山茱萸-白芍提取工艺研究

摘要
目的:采用星点设计-效应面法优化山茱萸-白芍最佳水提取工艺。方法:以莫诺苷、马钱苷含量之和与芍药苷的含量为考察指标,单因素考察提取次数对提取工艺的研究,星点设计-效应面法考察加水量和提取时间对提取工艺的影响,对结果进行多元线性回归和二项式拟合,利用效应面法筛选提取工艺,再预测分析。结果:最佳水提取工艺为加水量11. 2倍,提取2次,每次81 min。最佳工艺验证结果与二项式拟合方程预测值偏差均小于1%。结论:采用星点设计-效应面法优化山茱萸-白芍最佳煎煮提取工艺,方法简便,预测性良好。
        Objective: To optimize the extraction process of Cornus officinalis Sieb-Paeonia lactiflora Pall couplet medicines by Central Composite Design-Response Surface Methodology. Methods: Taking the sum of the contents of Monoside and Loganin and Paeoniflorin as the evaluation indexes,the effects of extraction times on the extraction process were investigated by single factor,the effects of water addition and extraction time on the extraction process were investigated by Central Composite Design-Response Surface Methodology,the results were fitted by multiple linear regression and binomial fitting,and the extraction process was screened by the method of Response Surface,and then predicted and analyzed. Results: The optimum water extraction process was 11. 2 times the amount of water added,extracting twice,each time for 81 minutes. The deviation between the validation results of the optimum process and the predicted values of the binomial fitting equation is less than 1%. Conclusion: Central Composite Design-Response Surface Methodology was used to optimize the extraction process of Cornus officinalis Sieb-Paeonia lactiflora Pall couplet medicines. The method was simple and predictive.
