摘要
新生多肽结合复合体(Nascent polypeptide-associated complex,NAC)是由α和β两个亚基组成的异二聚体复合物。水稻新生多肽结合复合体α链基因家族共有3个基因,分别位于1、3号和5号染色体上。本研究根据公布的水稻基因组序列,通过RT-PCR方法克隆了粳稻品种中花11号中α-NAC家族的3个基因。经生物信息学分析发现,水稻α-NAC基因家族的氨基酸序列比较保守,包含NAC和UBA两个结构域,与玉米NAC基因家族的亲缘关系最近。Os1gNAC和Os3gNAC、Os5gNAC的相似性分别为67.4%、81.8%,Os3gNAC和Os5gNAC的相似性为69.5%。本研究结果为Osα-NAC蛋白纯化及蛋白结构生物学研究提供参考,为今后Osα-NAC基因家族的生物学功能研究奠定基础。
        Nascent polypeptide- associated complex(NAC) is a heterodimer consisting of α and β subunits. There are three members of α-NAC in rice genome,which are localized in chromosome 1,3 and 5 respectively. The full open reading frames of the three α-NAC genes were cloned by RT- PCR from Oryza sativa cv. Zhonghua 11. Further bioinformatics analysis revealed that the amino acid sequence of Osα-NAC was conserved inNAC and UBA domains,and showed the highest similarity to ZmNACs. Os1gNAC shared67. 4% and 81. 8% homology with Os3gNAC and Os5gNAC respectively,whereas Os3gNAC and Os5gNAC shared 69. 5% homology. These results would provide references for protein purification and structural biological study,and lay a foundation for functional analysis of Osα-NAC.
