广藿香与紫苏药对及其单味药挥发性成分GC-MS分析

摘要
目的研究中药广藿香及紫苏挥发性成分在配伍后的含量及种类变化情况。方法参照国家药典,对广藿香和紫苏药对的单味药和药对挥发油都采用水蒸气蒸馏法提取,然后以气相色谱-质谱联用技术(GC-MS)分别对各挥发油成分进行分析,并且经计算机检索及质谱图解析确定化合物,最后采用峰面积归一法计算相对百分含量。结果发现广藿香和紫苏药对中新增了蘑菇醇、金合欢烯、γ-广藿香烯等8个化合物;广藿香中的α-石竹烯、α-广藿香烯、广藿香烯等10个化合物在药对中并未检测到;而紫苏中的紫苏醛、甲基丁香酚、异植物醇等9个化合物也没有在药对中检测到。此外,单味药中的α-蒎烯、β-蒎烯、广藿香醇等化合物的含量在药对中明显提高,而单味药中的石竹烯、广藿香酮等化合物含量则在药对中有所降低。结论两个单味药配伍导致其化学成分的种类和含量发生了较为明显的变化。
        Objective To study the variation of volatile components of Pogostemonis herba and Perillae folium after compatibility. Methods According to Chinese Pharmacopoeia, the volatile oil was extracted by steam distillation(SD), and components of each volatile oil were analyzed by gas chromatography-mass spectrometry(GC-MS).Compounds were determined by computer search and analysis of mass spectrograms,and the peak area normalization method was used to calculate the relative percentages. Results the experimental data showed that a total of 8 compounds including 1-Octen-3-ol, β-Farnesene and γ-Patchoulene, etc, were appeared in the herbal pair.There were 10 compounds in Pogostemonis herba that were not found in herbal pair, including Humulene, α-Patchoulene and Patchoulene,etc. In addition,9 compounds including Perillaldehyde,Methyleugenol and Isophytol in Perillae folium,were not detected in the herbal pair. And,the contents of compounds in single herb such as α-Pinene,β-Pinene,Patchouli alcohol,were significantly increased;while the contents of compounds in single herb such as Caryophyllene and Pogostone were decreased. Conclusion The chemical composition and contents changed after compatibility.
