摘要
目的:探讨附子炮制品炮天雄对急性心力衰竭大鼠血流动力学和神经体液因子的影响。方法:对健康成年大鼠采用左心室插管方法监测其心室内压及频率,盐酸普罗帕酮复制急性心力衰竭模型,造模成功后,分别对各组大鼠给药:模型组十二指肠给予蒸馏水,阳性组股静脉注射参附注射液(3.33 mL/kg),炮天雄高、中、低剂量组分别经十二指肠给予炮天雄水煎液5,2.5,1.25 g生药/kg。记录造模前后以及给药后5 min、10 min、20 min、30 min、60 min时心脏频率(HR)和左心室内压最大上升速率(+dp/dt_(max))、左心室内压最大下降速率(-dp/dt_(max))的变化。观察60 min后腹主动脉取血,采用酶联免疫法测定血清中Ang-I、ALD、ET-1、ANP和TNF-α含量。结果:不同剂量的炮天雄可不同程度的升高模型大鼠各时间点的HR和+dp/dt_(max),降低-dp/dt_(max);并能降低模型大鼠血清中Ang-I、ALD、ET-1、ANP、TNF-α水平。结论:炮天雄对盐酸普罗帕酮诱导的急性心力衰竭大鼠具有强心作用,能够改善其血流动力学,降低与心力衰竭有关的神经体液因子的分泌。
        Objective:To study the effects of processed Tianxiong on hemodynamics and neurohumoral factors of rats with acute heart failure(AHF).Methods:We used propafenone hydrochloride to copy the model rats of AHF.After it,we recorded the HR and left ventricular pressure by BL-420 system.Then,the model groups were given distilled water by the duodenum.The positive groups were injected Shenfu Injection(3.33 mL/kg),and the treatment groups were given processed Tianxiong with high,moderate and low doses(5,2.5,1.25 g crude drug/kg) by the duodenum.We observed and recorded the changes of HR,+dp/dt_(max) and-dp/dt_(max) after administration 5,10,20,30,60 min,and determined the concentrations of Ang-I,ALD,ET-1,ANP and TNF-α by blood sampling after 60 min by ELISA.Results:The different doses of processed Tiaoxiong can increase the HR,+dp/dt_(max) and decrease the-dp/dt_(max) of the rats with AHF in varying degrees at all points of time.And it also can decrease the concentrations of Ang-I,ALD,ET-1,ANP and TNF-α,which were increased by modeling.Conclusion:Processed Tiaoxiong has cardiac effect on rats with AHF,and can improve hemodynamic conditions and affect the neurohumoral factors associated with heart failure.
