滋阴养血安神方联合针刺对PCPA诱导的失眠小鼠肝脏TNF-α表达影响

摘要
目的 :初步探讨滋阴养血安神方联合针刺对PCPA诱导的失眠小鼠肝脏TNF-α表达的影响。方法:利用腹腔注射对氯苯丙氨酸(PCPA)制造睡眠剥夺模型,分5组,分别为空白对照组、模型组、针刺组(百会穴、神庭穴)、滋阴养血安神方组(中药组)、滋阴养血安神方联合针刺组(针药联合组),造模并给药7 d后,检测小鼠肝TNF-α的mRNA及蛋白表达。结果 :针刺组降低了模型组TNF-α蛋白表达,针药联合组显著降低了TNF-α蛋白表达(P<0.05),针药联合组效果更好。针刺组、针药联合组降低了TNF-α的mRNA表达水平,尤其是针药联合组显著降低了TNF-α的mRNA表达水平(P<0.01),针刺及滋阴养血安神方对降低肝脏TNF-α表达有一定的协同作用。结论 :滋阴养血安神方及针刺法可以影响PCPA诱导的失眠小鼠肝脏TNF-α表达,且针刺与中药具有一定的协同作用。
        Objective :To explore the effect of Ziyin Yangxue Anshen Decoction combined with acupuncture on TNF-α expression in the liver of PCPA-induced insomnia mice. Methods :Using the abdominal cavity injection of chlorobenzene alanine(PCPA)to manufacture sleep deprivation model,then 5 group could be devided,blank control group,model group,acupuncture group(Baihui,Shenting),Ziyin Yangxue Anshen Decoction group,Ziyin Yangxue Anshen Decoction combined acupuncture group,after building and delivery 7 days,detection of mice liver TNF-α expression in mRNA and protein level. Results :The acupuncture group reduced the protein level of TNF-α in the model group,especially the acupuncture group and the acupuncture binding drug group significantly reduced the level of TNF-α(P<0.05). The mRNA expression levels of TNF-α were decreased both in the acupuncture group and the combination group,especially in the acupuncture group and the combination group(P<0.01). Acupuncture and Ziyin Yangxue Anshen Decoction had certain synergistic effects on decreasing the expression of TNF-α in liver. Conclusion :Ziyin Yangxue Anshen Decoction combined acupuncture can affect the expression of TNF-α in the liver of insomnia mice induced by PCPA. Synergy of acupuncture and prescription can be seen.
