摘要
目的建立测定肠炎灌肠液中盐酸小檗碱含量的高效液相色谱(HPLC)法。方法色谱柱为Kromasil 100-5 C18柱(250 mm×4. 6 mm,5μm),流动相为乙腈-0. 1%磷酸溶液(50∶50,V/V,每100 m L加十二烷基磺酸钠0. 1 g),流速为1 m L/min,检测波长为265 nm,柱温为40℃,进样量为10μL。结果盐酸小檗碱进样质量浓度在6. 72~215. 02 g/L范围内与峰面积线性关系良好,精密度、稳定性、重复性试验结果的RSD均小于2%,平均加样回收率为101. 91%,RSD为1. 53%(n=6)。结论该方法准确、灵敏,可用于肠炎灌肠液的质量控制。
Objective To establish an HPLC method for the content determination of berberine hydrochloride in enterocolenteral enema.Methods The chromatographic column was Kromasil 100-5 C18 column( 250 mm × 4. 6 mm,5 μm),the mobile phase was acetonitrile-0. 1% phosphoric acid solution( 50 ∶ 50,V/V,0. 1 g sodium dodecylsulphate was added into the 100 m L mobile phase),the flow rate was1 m L/min,the detection wavelength was 265 nm,the column temperature was 40 ℃ and the sample size was 10 μL. Results Berberine hydrochloride showed good relationship in the range of 6. 72-215. 02 g/L. RSDs of precision,repeatability and stability tests were less than 2%,the average recovery rate was 101. 91%,RSD was 1. 53%( n = 6). Conclusion The method is accurate,sensitive and can be used for the quality control of enterocolenteral enema.
