摘要
目的:探究扶正抑瘤汤对人肝癌细胞Hu-7移植瘤裸鼠模型的影响。方法:对75只裸鼠进行人肝癌细胞Hu-7移植建模后,随机分成对照组、5-氟尿嘧啶组、高剂量扶正抑瘤汤组、中剂量扶正抑瘤汤组、低剂量扶正抑瘤汤组,分别给予生理盐水、5-氟尿嘧啶、扶正抑瘤汤(高、中、低剂量)干预3周后处死,记录和比较5组裸鼠的移植瘤质量、抑瘤率、凋亡细胞数量、自噬小体数量和Beclin-1、LC3、Bnip3表达情况。结果:5-氟尿嘧啶组、高剂量扶正抑瘤汤组、中剂量扶正抑瘤汤组、低剂量扶正抑瘤汤组裸鼠的肝癌移植瘤平均质量均低于对照组、抑瘤率均高于对照组(P<0.05);与中剂量扶正抑瘤汤组和低剂量扶正抑瘤汤组比较,5-氟尿嘧啶组、高剂量扶正抑瘤汤组裸鼠的肝癌移植瘤平均质量均降低,抑瘤率增加(P<0.05);电镜下,5-氟尿嘧啶组、高剂量扶正抑瘤汤组裸鼠的移植瘤细胞双层膜自噬小体数量较多,而中剂量扶正抑瘤汤组、低剂量扶正抑瘤汤组裸鼠的移植瘤细胞自噬小体较少;与对照组比较,5-氟尿嘧啶组、高剂量扶正抑瘤汤组、中剂量扶正抑瘤汤组的裸鼠的Beclin-1、LC3、Bnip3的表达增加(P<0.05),与中剂量扶正抑瘤汤组和低剂量扶正抑瘤汤组比较,5-氟尿嘧啶组、高剂量扶正抑瘤汤组的裸鼠的Beclin-1、LC3、Bnip3的表达增加(P<0.05)。结论:扶正抑瘤汤能抑制人肝癌细胞Hu-7移植瘤裸鼠肿瘤的生长,增强肿瘤细胞的自噬活性,以高剂量扶正抑瘤汤的作用最明显。
Objective:To investigate the effect of Fuzheng Yiliu Decoction(扶正抑瘤汤) on nude mice models of human hepatoma cell Hu-7 xenograft.Methods:Human hepatoma cell line Hu-7 was transplanted into 75 nude mice,which were randomly divided into control group,5-fluorouracil group and high dose group,medium dose group,low dose group,were given saline,5-fluorouracil,Fuzheng anti tumor decoction(high,medium and low dose) after 3 weeks of treatment were recorded and compared the transplanted tumor weight,inhibition rate,the number of apoptotic cells,the number of autophagosomes,the expression of Beclin-1,LC3 and Bnip3 between five groups.Results:Compared with the control group,the average weight of transplanted liver tumor of 5-fluorouracil group and high dose group,medium dose group,low dose group of nude mice were decreased and the inhibition rate increased(P<0.05);compared with medium dose group,Low dose group,the average weight of transplanted liver tumor 5-fluorouracil group and High dose Decoction group were decreased and the inhibition rate increased(P<0.05);under electron microscope,in the 5-fluorouracil group and the High dose group,the number of autophagic cells in the bilayer membrane of the transplanted tumor cells were higher;compared with the Control group,expression of Beclin-1,LC3,Bnip3 of 5-fluorouracil group and High dose group,Medium dose group were increased(P<0.05),compared with Medium dose group and Low dose group,expression of Beclin-1,LC3,Bnip3 of 5-fluorouracil group and High dose group were increased(P <0.05).Conclusion:Fuzheng Yiliu decoction can inhibit the growth of tumor cells in nude mice bearing human hepatoma cell Hu-7 and enhance the autophagic activity of tumor cells,and the effect of high dose Fuzheng anti tumor Decoction is most obvious.
