摘要
【目的】观察槲皮素对人骨肉瘤MG-63细胞体外生长周期及自噬的影响。【方法】以不同质量浓度的槲皮素作用于体外培养的MG-63细胞,流式细胞术检测MG-63细胞生长周期,荧光显微镜观察单体红色荧光蛋白(mRFP)—绿色荧光蛋白(GFP)—微管相关蛋白轻链3(LC3)双标腺病毒转染MG-63细胞后自噬小体的形成和自噬流的变化。【结果】槲皮素显著增加处于G1期的骨肉瘤MG-63细胞数量,上调MG-63细胞自噬水平。【结论】槲皮素可抑制人骨肉瘤MG-63细胞的增殖,使其停滞在细胞周期G1期,并诱导细胞自噬。
        Objective To investigate the effect of quercetin on in-vitro cell cycle and autophagy of human osteosarcoma MG-63 cells. Methods Human osteosarcoma MG-63 cells were cultured by different mass concentrations of quercetin in vitro. The cell cycle of MG-63 cells was measured by flow cytometry(FCM),and the63 cells transfected by monomeric red fluorescent protein(mRFP)-green fluorescent protein(GFP)-microtubule-associated protein 1 light chain 3(LC3)adenovirus were observed under fluorescence microscope. Results Quercetin significantly increased the proportion of MG-63 cells in G1 phase,and raised the level of autophagy of MG-63 cells. Conclusion Quercetin can inhibit the proliferation of human osteosarcoma MG-63 cells,arrest the cells restrain in G1 phase,and induce autophagy.
