摘要
目的:探讨淫羊藿次苷Ⅱ(icarisideⅡ,ICS-Ⅱ)对气管切开插管留置大鼠肺泡灌洗液分泌型免疫球蛋白A(secretory immunoglobulinA,sIgA)、肺组织β-防御素-2(rat Beta-defensin-2,rBD2)和外周血白细胞介素-6(interleukin-6,IL-6)含量的影响。方法:将54只SPF级雄性SD大鼠随机分成3组,正常对照组(A)、模型对照组(B)及用药组(C),每组18只。制备大鼠气管切开插管留置模型。模型成功6 h后给予C组ICS-Ⅱ[30 mg·(kg·d)~(-1)]灌胃,A、B组灌胃等量生理盐水。分别于24 h、72 h、168 h 3个时间段取外周血、肺泡灌洗液及肺组织,采用酶联免疫吸附法(ELISA)检测肺泡灌洗液sIgA及外周血IL-6含量,PCR检测肺组织rBD2 mRNA相对表达量。结果:气管切开后肺泡灌洗液sIgA含量24 h即明显升高(P<0.01),但在72 h后又降至正常水平;用ICS-Ⅱ干预后肺泡灌洗液sIgA含量在3个时间段均明显升高(P<0.05),以72 h升高最明显。气管切开插管后大鼠外周血清IL-6含量72 h明显降低(P<0.01),在24 h和168 h没有明显变化;用ICS-Ⅱ干预后,IL-6含量在3个时间段均明显下降(P<0.05)。气管切开插管后24 h rBD2表达明显增加(P<0.01),随后72 h、168 h时间段逐渐降低;用ICS-Ⅱ干预后,rBD2表达量没有升高,反而明显下降(P<0.05)。结论:ICS-Ⅱ能够提高气管切开插管大鼠sIgA分泌,改善肺部局部免疫功能。
Objective:To investigate the effect of icariside Ⅱ(ICS-Ⅱ) on tracheal intubation in rats with secretory immunoglobulin A(sIgA) and lung tissue β-defense,Beta-2(rBD2) and peripheral blood interleukin-6(IL-6).Methods:54 male SPF rats were randomly divided into 3 groups:normal control group(A),model control group(B) and model medication group(C),with 18 rats in each group.A rat tracheotomy cannula indwelling model was prepared.After 6 hours of successful model,group C was given ICS-Ⅱ [30 mg·(kg·d)~(-1)],and group A and B were given normal saline.Peripheral blood,alveolar lavage fluid and lung tissue were obtained at 24 h,72 h and 168 h,respectively.Enzyme-linked immune-sorbent assay(ELISA) was used to detect sIgA and peripheral blood IL-6 in alveolar lavage fluid.PCR The relative expression of rBD2 mRNA in lung tissue was detected.RESULTS:The content of sIgA in alveolar lavage fluid was significantly increased at 24 h after tracheotomy(P<0.01),but decreased to the normal level after 72 h.The sIgA content in alveolar lavage fluid after ICS-Ⅱ intervention It was significantly increased in all three time periods(both P<0.05),and the highest was observed at 72 h.After tracheotomy,the serum IL-6 level in rats was significantly decreased at 72 h(P<0.01),and there was no significant change at 24 h and 168 h.After ICS-Ⅱ intervention,IL-6 content was significantly decreased in three time periods(both P<0.05).The expression of rBD2 was significantly increased at 24 h after tracheotomy and intubation(P<0.01),and then gradually decreased at 72 h and 168 h.After ICS-Ⅱ intervention,the expression of rBD2 did not improve,but decreased significantly(both P<0.05).Conclusion:ICS-Ⅱ can improve the secretion of sIgA in tracheotomy and improve the local immune function of the lung.
