摘要
目的建立HPLC法同时测定金鸡虎补丸中去氢二异丁香酚、五味子甲素、染料木素和芒柄花素4种成分。方法金鸡虎补丸70%乙醇提取液的分析采用C18色谱柱(200mm×4.6mm,5μm);流动相A为乙腈-甲醇(3∶2),流动相B为0.1%H3PO4溶液,梯度洗脱;体积流量为0.9mL/min;去氢二异丁香酚检测波长为270nm,五味子甲素、染料木素和芒柄花素均为254nm;柱温为35℃;进样量20μL。结果被测定的4组成分色谱峰均有良好的分离度,方法精密度、重复性的RSD均小于2.0%;在室温条件下24h内稳定;各组分线性范围较宽,线性关系良好(r≥0.999 4);平均加样回收率为96.8%~98.1%,RSD(n=6)≤1.4%。结论金鸡虎补丸的提取方法和流动相保证了分析方法的简单易行、重复性好、准确可靠。
        AIM To develop an HPLC method for simultaneously determining four components,such as dehydrodiisoeugenol,deoxyschizandrin,genistein and formononetin in Jinji Hubu Pills.METHODS The chromatograhic separation of 70% ethanolic extract of Jinji Hubu Pills was performed on C18column( 200mm × 4.6mm,5 μm).Mobile phase consisted of acetonitrile-methanol( 3 ∶ 2)( A) and 0.1% phosphoric acid solution( B)were adopted at the flow rate of 0.9mL min.The detection wavelengths were 270nm for dehydrodiisoeugenol,254 nm for deoxyschizandrin,genistein and formononetin,respectively.The column temperature was 35 ℃.The injection volume was 20 μL.RESULTS The four compounds were well separated.The RSD values of precision and reproducibility were all less than 2%.The stability was good in 24h.The linear relationship between the concentration and peak areas of the four compounds was all greater than 0.999 4.The recoveries were 96.8%- 98.1%and RSD( n = 6) less than 1.4%.CONCLUSION The developed extraction and mobile phase were simple,sensitive,accurate,and reproducible for Jinji Hubu Pills.
