摘要
以朱顶红新生根尖为材料,采用常规压片法进行染色体制片,对朱顶红属品种进行了染色体核型分析及倍性鉴定。结果表明:0.002 mol/L 8-羟基喹啉水溶液4℃预处理朱顶红根尖9—24 h,1 mol/L HCl溶液60℃解离根尖9 min获得的染色体制片效果最好。首次对朱顶红品种'柠檬冰糕'进行核型分析,发现其为三倍体朱顶红,核型公式为K(2n)=3x=33=12 m+12 sm+9 st,核型不对称系数(As.K)为70.19%,核型类型为3B型。根据染色体制片结果对6个朱顶红品种进行倍性鉴定表明:'凤蝶'为二倍体朱顶红,'特伦蒂诺'为三倍体朱顶红,'粉色惊奇''快车''苹果花''樱桃妮芙'均为四倍体朱顶红。
Taking the new root tips as materials, karyotype analysis and ploidy identification of Hippeastrum cultivars were carried out by traditional chromosome tabletting method. The results showed that the best chromosome morphology was observed when root tips treated with 0. 002 mol/L 8-Hydroxyquinoline for 9—24 h at 4 ℃ and 1 mol/L HC1 for 9 min at 60 ℃. The karyotype of Hippeastrum cultivar ' Lemon Sorbet ' was analyzed for the first time. It was found that ' Lemon Sorbet ' was triploid,the karyotype formula was K(2 n)=3 x=33 =12 m +12 sm +9 st,the asymmetrical karyotype coefficient( As. K) was 70. 19% and the karyotype type was "3 B". With the same method,the ploidy identification of 6 Hippeastrum cultivars showed that H. papilio was diploid,' Trentino ' was triploid,' Pink Surprise ',' Rapido ',' Apple Blossom ' and ' Cherry Nymph ' were all tetraploid.
