意大利牛舌草抗氧化及抗血栓作用研究

摘要
目的研究意大利牛舌草水、乙醇提取物抗氧化及抗血栓的作用。方法将意大利牛舌草粉碎成粗粉,分别用蒸馏水和70%乙醇提取,得到意大利牛舌草水提取物和乙醇提取物。选取健康昆明小鼠70只,随机分为7组(n=10),正常对照组给予等体积蒸馏水,意大利牛舌草水提物高(678.40mg/kg)、中(339.20mg/kg)、低剂量组(169.60mg/kg),意大利牛舌草乙醇提物高(409.50mg/kg)、中(204.75mg/kg)、低剂量组(102.38mg/kg),各组小鼠每天灌胃给药1次,连续灌胃21d,按试剂盒方法说明书测定小鼠血清中的SOD、MDA、GSH-PX含量。测定意大利牛舌草水、乙醇提取物对正常小鼠凝血时间、止血时间及对正常大鼠动脉血栓形成的作用。结果与正常对照组比较,意大利牛舌草乙醇提物高剂量组小鼠血清中SOD、GSH-PX活性升高,MDA含量降低,差异有统计学意义(P <0.01);意大利牛舌草乙醇提物中剂量和低剂量组小鼠血清中SOD、GSH-PX活性升高,MDA含量降低,差异有统计学意义(P <0.05);意大利牛舌草水提物各剂量组小鼠血清中SOD、GSH-PX活性升高,MDA含量降低,差异无统计学意义(P>0.05)。意大利牛舌草乙醇提物(204.75mg/kg)和水提物(339.20mg/kg)均可延长正常小鼠的凝血时间,意大利牛舌草乙醇提物(141.75mg/kg)和水提物(234.83mg/kg)均可降低大鼠血栓湿重,血栓抑制率分别为42.76%、36.68%。结论意大利牛舌草乙醇提取物具有抗氧化作用,意大利牛舌草水、乙醇提取物可以延长凝血时间,防止血栓的形成。
        Objective To study the antioxidant and antithrombotic effects of water and ethanol extracts of Anchusa italica Retiz.Methods Anchusa italica Retiz.was crushed into crude powder and extracted with distilled water and 70% ethanol respectively.The water extract and ethanol extract of Anchusa italica Retiz.were obtained.70 healthy Kunming mice were randomly divided into 7groups(n =10).The control group was given distilled water.There were high-dose(678.40mg/kg),medium-dose(339.20mg/kg),low-dose(169.60 mg/kg)of water extraction groups,and high-dose(409.50 mg/kg),medium-dose(204.75mg/kg),and low-dose(102.38 mg/kg)of ethanol extraction groups.The dosage was given to mice for 21 days.The level of SOD,MDA and GSH-PX in the serum of mice were determined according to the kit instructions.The effects of water and ethanol extractions of Anchusa italica Retiz.on coagulation time,hemostasis time and arterial thrombosis in normal mice were examined.Results Compared with the control group,there were an increase on the level of SOD and GSH-PX and decrease on MDA in high dose ethanol extraction group,and the differences were statistically significant(P <0.01).The level of SOD and GSH-PX in serum of mice in medium-dose and low-dose of ethanol extraction group increased and the level of MDA decreased,and the differences were statistically significant(P <0.05).The level of SOD and GSH-PX in serum of mice from each dose group of water extraction groups were increased,and the content of MDA was decreased,but the difference was not statistically significant(P >0.05).Ethanol extraction(204.75mg/kg)and water extraction(339.20mg/kg)can significantly prolong the clotting time of blood.Both ethanol extraction(141.75mg/kg)and water extraction(234.83mg/kg)of Anchusa italica Retiz.could reduce the wet weight of thrombus in rats,and the thrombus inhibition rates were 42.76%and 36.68%,respectively.Conclusion The ethanol extract of Anchusa italica Retiz.has anti-oxidation effect,and the extraction of water and ethanol can significantly prolong the clotting time and prevent the thrombosis.
