中药有效部位组方通痹四味方的体内抗炎、镇痛及活血通络等药效评价

摘要
目的观察中药有效部位组方通痹四味方(TBSWF)体内抗炎镇痛、活血通络的药效,探究其作为风湿骨病候选组方的成药性。方法随机将昆明小鼠分为空白组,阿司匹林组(AP 250 mg·kg~(-1)),腰痛宁组(YTN220 mg·kg~(-1)),腰痹通组(YBT 560 mg·kg~(-1)),通痹四味方(TBSWF)高、中、低剂量组(50,100,200 mg·kg~(-1)),采用热致痛反应及醋酸致扭体反应模型考察组方的镇痛作用;采用二甲苯致耳廓肿胀模型考察组方的抗炎消肿作用;采用角叉菜胶诱发的足跖肿胀模型考察组方对急性关节炎的抑制效果;在以上7组的基础上再加入一组模型组,采用小鼠毛细血管通透性改变模型考察组方的活血通络作用。结果与空白组相比,二甲苯致耳廓肿胀模型TBSWF中剂量组和AP组肿胀率均显著降低(P<0.05,P<0.01);热致痛反应模型,TBSWF高剂量组在给药后0.5 h能够显著延长痛阈值(P<0.01);醋酸扭体实验中各组扭体次数均显著减少(P<0.01),其中TBSWF高、中剂量组的扭体次数均显著少于阿司匹林组及YTN、YBT组(P<0.01);角叉菜胶诱发的足跖肿胀实验中TBSWF方高、中剂量组分别在给药后1 h和2~3 h肿胀度显著降低(P<0.05,P<0.01)。小鼠毛细血管通透性实验中AP及TBSWF各剂量组的吸光度均低于模型组(P<0.01)。结论 TBSWF在抗炎、镇痛、活血通络及小鼠足肿胀急性关节炎模型下的表现均优于腰痛宁胶囊和腰痹通胶囊,并且各模型下都至少有一个TBSWF给药组与阿司匹林效果相当,TBSWF对小鼠醋酸扭体的镇痛效果显著优于3个阳性对照药。通痹四味方可作为治疗风湿骨病的候选中药复方。
        Objective To explore the possibility of Tongbi Siwei Formula(TBSWF),a formula composed of traditional Chinese medicine effective parts,to be a candidate of traditional Chinese formula for curing rheumatoid bone diseases by observing its in-vivo anti- inflammatory,analgesic,and blood- activating and collateral- dredging effects. Methods Kunming mice were randomly divided into seven groups,namely blank control group,aspirin group(250 mg·kg-1),Yaotongning group(YTN,220 mg·kg~(-1)),Yaobitong group(YBT,560 mg·kg~(-1)),and low-,medium- and high- dose TBSWF groups(50, 100, 200 mg·kg-1). Hot- plate- induced pain response model and acetic- acid- induced writhing model were applied to investigate the analgesic effect, xylene- induced auricular edema model was used to study the anti- inflammatory effect, carrageenan- induced mice paw edema model was used to examine the effects on treating acute arthritis. By the establishment of a model group,mice capillary permeability test was adopted for the observation of blood- activating and collateral- dredging effects. Results Compared with the blank control group, aspirin and middle- dose TBSWF reduced the auricular swelling rate of xylene- induced ear edema animal model(P<0.01),and high- dose TBSWF increased the mice pain threshold 0.5 hour after medication in hot- plate- induced pain response test(P < 0.01); in acetic- acid- induced writhing experiment, the writhing frequency of all medicated groups was significantly reduced(P < 0.01),and the frequency in high- and middle- dose TBSWF groups was lower than that of aspirin group, YTN group and YBT group(P < 0.01); in carrageenan- induced paw edema experiment, swelling degree of high- and middle- dose TBSWF groups was reduced obviously one hour or 2- 3 hours after medication(P <0.01);in mice capillary permeability test,the absorbance of aspirin group and all TBSWF groups were significantly dropped(P < 0.01). Conclusion TBSWF has better anti- inflammatory, analgesic, blood- activating and collateraldredging,and anti- acute- arthritis effects than YTN and YBT,and TBSWF at certain dose in each experiment shows the effects similar to those of aspirin. Moreover,the analgesic effect of TBSWF on acetic- acid- induced writhing mice is significantly superior to that of the three positive control drugs. Therefore,it is feasible that TBSWF will be used as a candidate of traditional Chinese formula for curing rheumatoid bone diseases.
