摘要
为了建立一种有效的条浒苔细胞壁破壁方法,以显微观察和蛋白质提取率为评价指标,考察干法超微粉碎法、湿法超微粉碎法、超声波粉碎法、湿法超微粉碎+酶解法和超声波粉碎+酶解法5种方法对条浒苔细胞壁的破坏效果.结果表明,5种方法中湿法超微粉碎+酶解法处理后,细胞组织形态被破坏,细胞之间排布零散无规律,细胞内组织液流出,此方法蛋白质提取率最高,可达到30.86%.
In order to establish an effective cell wall breaking method for Enteromorpha clathrata,five methods which include dry superfine pulverization,wet ultrafine pulverization,ultrasonic pulverization,ultrasonic pulverization with enzymatic hydrolysis and wet ultrafine pulverization with enzymatic hydrolysis are investigated to evaluate the effect of destruction on the cell wall of Enteromorpha clathrata by using microscopic observation and the protein extraction rate as index.The results demonstrate that the method of wet ultrafine pulverization with enzymatic hydrolysis treatment can breakdown the cell morphous,make the cells scatter irregularly and the enchylema is flowed out.The extraction rate of protein treated by this method can reach 30.86%,which is the highest among the tested methods.
