啤酒灌胃对糖尿病大鼠血糖代谢功能影响的实验研究

摘要
目的探讨饮用啤酒对2型糖尿病(DM)大鼠血糖代谢功能的影响。方法 72只6月龄SPF级雄性Wistar大鼠成功建立DM大鼠模型,随机分为模型组(等量蒸馏水灌胃)、低剂量组(0.5 g·kg~(-1)·d~(-1)啤酒灌胃)、中剂量组(2.5 g·kg~(-1)·d~(-1)啤酒灌胃)、高剂量组(5 g·kg~(-1)·d~(-1)啤酒灌胃)各18只,另选18只正常大鼠作为正常组(等量蒸馏水灌胃),灌胃时间均为8周。末次灌胃后取尾静脉血测量不同时间点各组大鼠血糖、胰岛素、胰岛素敏感指数(ISI)、胰高血糖素样肽-1(GLP-1)。结果灌胃前(0 min)时,模型组、低剂量组、中剂量组、高剂量组的血糖、胰岛素、GLP-1水平显著的高于正常组(P<0.05);末次灌胃后(30 min、60 min、90 min、120 min、180 min)时,中剂量组、高剂量组的血糖、胰岛素水平高于正常组、模型组和低剂量组(P<0.05),GLP-1水平显著低于模型组和低剂量组(P<0.05);低剂量组、模型组的血糖、胰岛素、GLP-1水平在末次灌胃后(30 min、60 min、90 min、120 min、180 min、240 min)时比较差异均无统计学意义(P>0.05);灌胃前(0 min)时,模型组、低剂量组、中剂量组、高剂量组的ISI测定值显著低于正常组(P<0.05);末次灌胃后(30 min、60 min、90 min、120 min、180 min、240 min)时,中剂量组、高剂量组的ISI水平低于正常组、模型组和低剂量组(P<0.05);低剂量组、模型组的ISI水平在末次灌胃后(30 min、60 min、90 min、120min、180 min、240 min)时比较差异均无统计学意义(P>0.05)。结论少量啤酒对DM大鼠血糖代谢影响不显著,增加饮酒剂量会降低大鼠胰岛素敏感度及GLP-1水平,进一步升高血糖水平。
        Objective To investigate the effects of beer on glucose metabolism in type 2 diabetes mellitus( DM) rats. Methods 72 SPF male Wistar rats of 6 months which were successfully established DM rat model were randomly divided into model group( distilled water gavage),low dose group( 0. 5 g· kg~(-1)·d~(-1) beer gavage),middle dose group( 2. 5 g· kg~(-1)·d~(-1) beer gavage),high dose group( 5 g· kg~(-1)·d~(-1) beer gavage). 18 rats in each group and another 18 rats were in normal control group( distilled water gavage). The intragastric administration lasted for 8 weeks. The blood glucose,insulin,insulin sensitivity index( ISI) and glucagon like peptide-1( GLP-1) were measured at different time points after the last intragastric administration. Results Before gavage( 0 min),blood glucose,insulin and GLP-1 levels of model group,low dose group,middle dose group and high dose group were significantly higher than that of normal group( P < 0. 05). Blood glucose and insulin levels of middle dose group and high dose group in last intragastric administration( 30 min,60 min,90 min,120 min,180 min) were higher than those of normal group,model group and low dose group( P < 0. 05). GLP-1 of model group and low dose group were significantly lower than those of low dose group,model group( P < 0. 05). There was no statistically significant difference between blood glucose,insulin levels,GLP-1 after intragastric administration( 30 min,60 min,90 min,120 min,180 min,240 min) in low dose group and model group( P > 0. 05). ISI value of model group,low dose group,middle dose group and high dose group before gavage( 0 min) were significantly lower than those of normal group( P < 0. 05). The ISI of middle and high dose group after intragastric administration( 30 min,60 min,90 min,120 min,180 min,240 min) were lower than those of normal group,model group and low dose group( P < 0. 05). There was no significant difference of ISI in low dose group,model group after intragastric administration( 30 min,60 min,90 min,120 min,180 min,240 min)( P > 0. 05). Conclusion A small amount of beer has no significant influence on blood glucose metabolism in DM rats,and increased drinking dose can decrease insulin sensitivity and GLP-1 levels,and further increase blood glucose levels.
