摘要
目的:优化牛大力总黄酮提取工艺。方法:采用超声提取法,以总黄酮提取率为评价指标,以乙醇浓度、乙醇体积、提取时间为考察因素,利用Box-Behnken效应面法优化提取工艺。结果:最佳工艺为乙醇浓度81.07%、乙醇体积21.63倍、超声提取2次、每次52.51 min,温度60℃。在此条件下,牛大力总黄酮提取率达3.81 mg/g。结论:Box-Behnken效应面法优选的提取工艺稳定可行,实验值与预测值相吻合。
Objective To optimize extraction technology of totals flavonoids from Radix Millettia speciosa. Method On the basis of single factor experiment, ultrasonic extraction method was adopted. Ethanol concentration, solvent volume and extraction time were used to examine the yield of total flavonoids by Box-Behnken response surface methodology. Result The optimum conditions of extraction process consisted of 81.07% ethanol for 21.63 times, 52.51 min extraction time, and twice ultrasonic with 60 ℃, while the extraction rate of totals flavonoids was 3.81 mg/g. Conclusion This optimized technology was stable and feasible by Box-Behnken response surface methodology, which the predictive value and the experimental observation value was fitted.
