摘要
目的:该研究旨在观察中药复方芪丹地黄颗粒(QDDHG)对糖尿病并发症的治疗作用。方法:将SD大鼠腹腔注射链脲佐菌素(STZ)诱导成糖尿病模型,实验分为6组(每组n=9),即正常对照组,糖尿病模型组,QDDHG高、中、低剂量组,及阳性药物(氯沙坦)对照组。用氯沙坦或QDDHG治疗4周后,收集血液样品。分别于给药0、2、4、6、8、10周检测血糖和体质量。给药后第10周处死大鼠采血检测其血脂和血清肌酐(Scr)指标。前一天收集大鼠24 h尿液(24U-vol),记录尿量,并检测24 h尿蛋白(24 hU-pro)、内生肌酐清除率(Ccr),取器官进行病理形态学检测。结果:通过QDDHG治疗可显著改善糖尿病大鼠Scr、Ccr,低密度脂蛋白(LDL)及甘油三酯(TG),与正常组比较(P <0. 05)。对24 h U-pro改善尤为明显(P <0. 01),对血糖和体质量则无明显影响。病理结果显示对糖尿病大鼠的改善作用主要在肾脏,对胰腺,心脏和肝脏则没有明显影响。结论:QDDHG具有肾脏保护作用与血脂的调节功能。
        Objective:The purpose of this study was to observe the therapeutic effect of Chinese herbal compound Qidan Dihuang Granule( QDDHG) on diabetic complications. Methods:SD rats were intraperitoneally injected with streptozotocin( STZ) to induce diabetes. The experiment was divided into 6 groups( n = 9 in each group),including normal control group,diabetes model group,QDDHG high,medium and low dose groups,and a positive drug( losartan) control group. After 4 weeks treatment with losartan or QDDHG,blood samples were collected. Blood glucose and body weight were measured at 0,2,4,6,8 and 10 weeks,respectively. At the 10 th week after the administration,the rats were sacrificed for blood sampling and serum creatinine( Scr). The rats were collected 24 h urine(24 U-vol) the day before,and the urine volume was recorded. The 24-hour urine protein(24 hU-pro) and endogenous creatinine clearance rate( Ccr) were detected,and the organs were taken out for pathological examination. Results:Scr,Ccr,low-density lipoprotein( LDL) and triglyceride( TG) in diabetic rats were significantly improved by QDDHG treatment compared with the normal group( P < 0. 05). The improvement of 24 h U-pro was especially significant( P <0. 01),while it had no significant effect on blood glucose and body weight. Pathological results showed that the improvement in diabetic rats was mainly in the kidneys,but there was no significant effect on the pancreas,heart or liver. Conclusion:QDDHG has renal protective effect and regulation of blood lipids.
