摘要
以‘新球蜜荔’和‘大丁香’2个荔枝品种胚性愈伤组织为材料,研究了‘新球蜜荔’(XQTD)和‘大丁香’(DDXTD)接种在TD(MS+0.1 mg/L NAA+0.5 mg/L TDZ+0.1 g/L肌醇+0.4 g/L水解乳蛋白(LH)+100 mL/L椰汁+60g/L蔗糖+10 g/L琼脂)培养基与‘新球蜜荔’接种在TX(MS+0.1 mg/L NAA+5 mg/L ZT+0.1 g/L肌醇+0.4 g/L水解乳蛋白(LH)+100 ml/L椰汁+60 g/L蔗糖+10 g/L琼脂)培养基(XQTX)上各培养物体胚发生过程中抗氧化酶与多胺氧化酶活性变化。结果表明:体胚发生过程中,三者的可溶性蛋白含量变化都呈单峰曲线,峰值出现在第6天,在前12 d的培养中,XQTD高于XQTX,仅在第9天,DDXTD高于XQTD。三者的丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性都变化平缓且差异不大。三者的过氧化物酶(POD)活性变化都呈单峰曲线,XQTD的峰值出现在第9天,其余出现在第12天,体胚发生过程中DDXTD最高。XQTD和XQTX的过氧化氢酶(CAT)活性都呈先降后升趋势,而DDXTD只是后期下降。三者的多胺氧化酶(PAO)活性变化都呈先降后升趋势,在前15 d的培养中,XQTX和DDXTD都低于XQTD,且DDXTD在前6 d里基本无变化。培养物中可溶性蛋白含量、POD和PAO活性变化可能与‘新球蜜荔’及‘大丁香’的体胚发生效率差异有关。
        Changes of several antioxidant enzymes and polyamine oxidase activity in embryonic callus of litchi cultivars ‘Xinqiumili' and ‘Dadingxiang' were studied during the somatic embryogenesis induced on different induction media.The results showed that there was one peak curve of soluble protein content in both the embryonic callus of ‘Dadingxaing'(DDXTD) and ‘Xingqiumili'(XQTD)inoculated on TD medium(MS + 0.1 mg/L NAA + 0.5 mg/L TDZ + 0.1 g/L inositol + 0.4 g/L LH + 100 ml/L coconut juice + 60 g/L sucrose + 10 g/L agar), and the embryonic callus of ‘Xingqiumili'(XQTX)inoculated on TX medium(MS + 0.1 mg/L NAA + 5 mg/L ZT + 0.1 g/L inositol + 0.4 g/L LH + 100 ml/L coconut juice + 60 g/L sucrose + 10 g/L agar), with the peak occurring on the 6 th day after inoculation(th day). The content of soluble protein in XQTD was higher than that in XQTX from 6 th to 12 th day, while DDXTD had higher soluble protein content on 9 th day than XQTD. The malondialdehyde(MDA) content and the superoxide dismutase(SOD) activity in XQTX, XQTD and DDXTD had gentle variations and little differences. During the process of somatic embryogenesis, the changes of peroxidase(POD) activity in XQTX, XQTD and DDXTD showed a single peak curve, with the peak of XQTD appeared on the 9 th day, the rest appeared on the 12 th day. The highest POD activity was in DDXTD, and the lowest was on XQTD. The catalase(CAT) activity in XQTD and XQTX increased after decrease during somatic embryogenesis, while the catalase(CAT) activity in DDXTD only decreased in the later period. The polyamine oxidase(PAO) activity in XQTD, XQTX and DDXTD all increased after a slight decrease in the early 6 days. PAO activity in XQTD was higher both than that in XQTX from the 6 th to 15 th day, and than that in DDXTD from the 0 to 15 th day. The soluble protein content, POD activity and PAO activity may be related to the different frequency of the somatic embryogenesis of 'Xinqiumili' and 'Dadingxiang'.
