益气解毒中药复方对4 Gy ~(60)Coγ射线致小鼠外周血细胞减少及免疫器官损伤的治疗效应研究

摘要
目的研究益气解毒中药复方对4 Gy~(60)Coγ射线致小鼠外周血细胞减少及免疫器官损伤的治疗效应。方法将120只Balb/c小鼠随机分为空白组、照射组,照射组小鼠以4 Gy~(60)Coγ射线照射,照后当天,将照射组随机分为模型组和中药组。照射后当天开始,空白组和模型组灌胃去离子水,中药组灌胃中药复方药液,每日1次,各组小鼠于照后第14、21、35、49 d分批次取材,每批小鼠均灌胃至取材前一天。检测外周血白细胞计数(WBC)、淋巴细胞计数(LYMP)、红细胞计数(RBC)、血红蛋白浓度(HGB)、血小板计数(PLT)、脾脏及胸腺指数。脾脏病理切片,HE染色观察脾脏组织形态学。结果与空白组比较,照后14 d,模型组小鼠外周血各类血细胞数及免疫脏器指数显著降低,脾脏病理改变明显;照后21 d,模型组各类血细胞数逐渐回升,脾脏指数恢复至正常水平,胸腺指数出现再次下降,脾脏病理改变有所减轻,切片显示红髓髓外造血明显;照后35 d,模型组胸腺指数恢复至正常水平,中药组RBC、HGB、PLT恢复至正常水平;照后49 d,模型组RBC、HGB、PLT方恢复至正常水平,WBC、LYMP仍低于空白组,脾脏结构接近正常,中药组WBC、LYMP已回升至正常水平。与模型组比较,照后14 d,中药组脾脏指数显著升高,脾脏病理改变较轻;照后35 d,中药组RBC和HGB显著升高;照后49 d,中药组WBC显著升高。结论益气解毒中药复方可促进照后小鼠外周血细胞和免疫器官的恢复,对急性辐射损伤小鼠外周血细胞减少及免疫器官损伤的治疗有一定疗效。
        Objective To study the treatment effect of a prescription of Chinese medicine which tonifies qi and removes toxicity on peripheral blood cytopenia and immune organs injury caused by 4 Gy~(60) Co γ ray in mice. Methods A total of 120 Balb/c mice were randomly divided into two groups:the blank group and the irradiation group which were followed by whole-body exposure to 4 Gy~(60) Co γ ray. After irradiation,the irradiated mice were randomly divided into two groups: the model group and the Chinese medicine group. Mice of the blank and model groups were given deionized water by gavage once daily,while the others were administrated with the Chinese medicine. On the 14 th,21 th,35 th,49 th day of exposure,the mice were sacrificed respectively and the white blood cell( WBC),lymphocytes( LYMP),red blood cell( RBC),hemoglobin( HGB),blood platelets( PLT),thymus index,spleen index and spleen pathological section of each group were measured. Results Compared with the blank group,the peripheral blood cell amount and immune organ index decreased significantly and the pathological changes of the spleen were obvious in the model group on 14 th day of exposure. Atday 21 after irradiation,all kinds of blood cells in the model group gradually increased,the spleen index returned to normal level,the thymus index decreased again,the pathological changes of the spleen were alleviated,and the pathological sections showed that the hematopoiesis was obvious outside the red pulp. At day 35 after irradiation,thymus index of the model group returned to normal level,and the RBC,HGB,PLT of the Chinese medicine groupregained to normal level. At day 49 after irradiation,the RBC,HGB,PLT of the model group just returned to normal level and the spleen structure was close to normal,while the WBC,LYMP were still lower than those of the blank group. The WBC,LYMP of the Chinese medicine group had returned to normal level. Compared with the model group,14 days after irradiation,the spleen index of the Chinese medicine group increased significantly,and the pathological changes of the spleen were slight; on the 35 th day after irradiation,the RBC and HGB of the Chinese medicine group were significantly increased; and the WBC of the Chinese medicine group significantly increased on the 49 th day after irradiation. Conclusion The prescription of Chinese medicine speeds up the recovery of peripheral blood cytopenia and immune organs injury after radiation in mice.
