略阳乌鸡内源性逆转录病毒鉴定及表达分析

摘要
内源性禽白血病病毒等以多种逆转录病毒基因组稳定存在鸡染色体中,其致病性低或无致病性,但是内源性病毒的基因表达降低了鸡抵抗外源病毒感染的能力。本研究以略阳乌鸡为研究对象,利用多重PCR鉴定鸡基因组中禽内源性病毒EAV-HP和禽白血病病毒E亚群(ALV-E),分析ALV-E的env基因表达情况。结果表明,30只略阳乌鸡的基因组中均携带EAV-HP病毒序列,其中28只携带ALV-E序列。通过MEGA和NETWORK软件对ALV-E的gp85基因分析表明,略阳乌鸡与江苏绿壳蛋鸡聚在同一分支,两者同源性高达99.1%。此研究也为后续略阳乌鸡遗传背景分析和选育抗病毒感染优良品种提供基础数据。
        Endogenous avian leukosis virus and others as retroviral genomes stablely present in chicken chromosomes,and their pathogenicity is low or no pathogenicity.However,endogenous viral gene expression reduces the ability of chicken to resist exogenous virus infection.This study took Lueyang black-bone chicken as a research object,EAV-HP and avian leukemia virus ALV-E of Lueyang black-bone chicken genome were identified by multiple PCR,and the expression of env gene in ALV-E was analyzed.The results demonstrated that EAV-HP virus sequences were found in genomes of 30 Lueyang black-bone chickens,and 28 of 30 chickens harbored ALV-E virus sequence.The results of gp85 sequences analysis via MEGA and NETWORK softwares illustrated that Lueyang black-bone chicken and Jiangsu green eggshell chicken were clustered in the same clade,with homology up to 99.1%.This study provided the basic data for the subsequent analysis of genetic background of Lueyang black-bone chicken and selection of good breeds with antiviral ability.
