摘要
目的葛花中总黄酮含量的测定及提取工艺的优化。方法以总黄酮提取率为考察指标,采用冷凝回流、分光光度检测的方法,考察了单因素乙醇体积分数、物料比、提取时间对提取率的影响,采用3因素4水平[L16(43)]的正交实验,优化了提取工艺。结果最佳提取工艺条件为:乙醇体积分数为80%,物料比为1∶25(g∶m L),提取时间为1. 5h,提取4次,提取率为40. 54%。结论优化后的葛花总黄酮提取工艺合理可行,能为实际生产提供参考。
        Objective To establish the determination method and optimization of extraction process of total flavonoids from Pueraria lobata. Methods Using the orthogonal experimental design method involving three factors and four levels [L16( 43) ],and taking the volume fraction,material ratio and extraction time of ethanol as factors and the extraction rate of total flavonoids as the index,both wavelength scanning methed and condensation reflux were used to optimize the above three factors and determine the optimum extraction process. Results The optimum extraction conditions were as follows: optimum volume fraction of ethanol was 80%,optimum raio of Pueraria to ethanol was 1∶ 25,the optimum reaction time was 1. 5 h. Under the optimum extraction conditions,the average extraction rate of toal flavonoids was 40. 54%. Conclusion The establishment of determination method and extraction process of tatol flavonoids from Pueraria lobata is reasonable and feasible,which could provide theoretical reference for the establishment of novel procedures for actual production.
