山药块茎膨大过程内源激素的变化及相关基因的表达验证

摘要
山药(Dioscorea opposite, 2n=40)属于薯蓣科,其地下茎能膨大,膨大的地下茎具有良好的食用和药用价值,为了解析山药块茎膨大的分子机制,本研究对块茎膨大的3个时期对其叶片、地上茎、块茎各部位进行取样,测定内源激素含量,并结合山药转录组数据,对植物激素相关基因进行表达验证。研究表明:IAA、JA、MeJA全部呈上升趋势,而GA3含量先上升后下降;IAA相关基因SAUR (Unigene8864_All)和PIN domain-containing protein (CL3538.Contig1_All)在块茎膨大过程起促进作用;JA相关基因Allene oxide synthase (CL3684.Contig3_All)起促进作用。研究表明山药块茎膨大与植物激素息息相关,为深入研究山药块茎膨大进程,提高山药产量和质量提供了理论依据。
        Yam(Dioscorea opposite 2 n=40) belongs to Dioscorea family. Its underground stem can enlarge, which has good edible and medicinal value. In order to understand the molecular mechanism of yam tuber enlargement,this study sampled the leaves, aboveground stems and tubers at three stages of tuber enlargement, determined the level of endogenous hormones, and combined with the transcriptome data of yam to verify the related genes expression of plant hormones. The research showed IAA, JA and Me JA all tend to rise up while GA3 increased first and then decreased. IAA-related genes SAUR(Unigene 8864_All) and PIN domain-containing protein(CL35-38.Contig1_All) play a role in promoting tuber expansion. JA related gene allene oxide synthase(CL3684.Contig3_All) plays a promotive role. These results indicated that the expansion of yam tubers was highly related to plant hormones, which provided a theoretical evidence for further studying the process of tuber expansion and improving the yield and quality of yam.
