生巴戟天与盐巴戟天改善三氯化铝诱导的老年痴呆小鼠学习记忆能力的研究

摘要
目的比较生巴戟天与盐巴戟天对老年痴呆模型小鼠脑组织去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、超氧化物歧化酶(SOD)和丙二醛(MAD)的影响.方法将50只小鼠随机分为对照组、模型组、阳性对照组、生巴戟天组和盐巴戟天组.除对照组外,其余各组通过灌胃给予AlCl3(200mg·kg-1)建立老年痴呆模型,连续50d.生巴戟天组、盐巴戟天组小鼠分别灌胃给予6g·kg-1的生巴戟天、盐巴戟天水提物,阳性对照组小鼠灌胃给予32mg·mL-1吡拉西坦,对照组与模型组分别灌胃给予等体积的生理盐水.采用Morris水迷宫法评价学习记忆能力,采用高效液相色谱-电化学法检测小鼠脑组织中NE,DA和5-HT含量,采用考马斯亮蓝测定法测定小鼠脑组织中SOD和MAD活性.结果与对照组比较,模型组小鼠逃避潜伏期显著增加(P<0.05),穿越次数显著减少(P<0.05),脑组织NE,DA,5-HT含量与SOD活性显著下降(P<0.05),MAD活性显著升高(P<0.05);与模型组比较,阳性对照组、生巴戟天组、盐巴戟天组小鼠的逃避潜伏期显著减少(P<0.05),穿越次数显著增加(P<0.05);脑组织NE,DA,5-HT含量与SOD活性显著增高(P<0.05),MAD活性显著降低(P<0.05);盐巴戟天组小鼠脑组织NE,DA,5-HT含量与SOD活性明显高于生巴戟天组(P<0.05),MAD活性明显低于生巴戟天组(P<0.05).结论生巴戟天与盐巴戟天水提物均能提高抗氧化能力,提高单胺类神经递质含量,起到改善老年痴呆小鼠学习记忆能力.盐巴戟天对老年痴呆小鼠学习记忆能力能改善作用显著优于生巴戟天.
        Aims To investigate the effects of Morinda Officinalis and salted Morinda Officinalis on noradrenaline(NE),dopamine(DA),5-hydroxytryptamine(5-HT),superoxide dismutase(SOD)and malondialdehyde(MAD)in brain tissue of Alzheimer model mice.Methods Fifty mice were randomly divided into control group,model group,positive control group,Morinda Officinalis group and salted Morinda Officinalis group.In addition to the control group,the other groups were given AlCl3(200 mg·kg-1)by gavaging for 50 days to establish Alzheimer's disease model.Mice in Morinda Officinalis group and salted Morinda Officinalis group were given 6g·kg-1 of water extract of Morinda Officinalis group and salted Morinda Officinalis by intragastric,Mice in the positive control group were given 32mg·mL-1 piracetam by gavage.and the control group and the model group were given equal volume of normal saline.The study and memory ability were tested by Morris water maze method,and NE,DA,5-HT contents in the brain tissue were detected by HPLC-electrochemical method.SOD,MAD activity were detected by Coomassian lianglan assay.Results Compared with the control group,the escape latency in model group was significantly increased(P<0.05),crossing time was significantly decreased(P<0.05),and NE,DA,5-HT content and SOD activity in brain tissue were significantly decreased while MAD activity was significantly increased(P<0.05).Compared with model group,the escape latency in positive control group,Morinda Officinalis group and salted Morinda Officinalis group were significantly decreased(P<0.05),crossing time was significantly increased(P<0.05),and NE,DA,5-HT content and SOD activity in brain tissue were significantly increased while MAD activity was significantly decreased(P<0.05).The NE,DA,5-HT contents and SOD activity in brain tissue of salted Morinda Officinalis group were obviously higher than those in the Morinda Officinalis group(P<0.05),and MAD activity in brain tissue of salted Morinda Officinalis group were obviously lower than that in the Morinda Officinalis group(P <0.05)Conclusion Both Morinda Officinalis and salted Morinda Officinalis can improve the learning and memory ability of Alzheimer model mice by improving the antioxidant ability and increasing the content of monoamine neurotransmitters.The improvement of learning and memory ability of salted Morinda Officinalis in Alzheimer model mice was significantly better than that of Morinda Officinalis.
