摘要
目的探讨不同谷树叶提取物对高脂血症模型大鼠血脂水平及血管内皮细胞Bax和Bcl-2表达的影响。方法选取60只正常雄性SD大鼠,随机分为正常对照组、高脂血症模型对照组、辛伐他汀组及谷树叶不同提取物组(水提组、水提醇沉组和醇提组),每组10只;高脂饲料喂养8周造模,造模同时分别进行相应药物干预。分别于第4周和第8周检测大鼠血脂各项指标的水平,并检测血管内皮细胞Bax和Bcl-2的表达。结果与模型组大鼠比较,谷树叶各提取物组的TC和LDL-C水平均降低,HDL-C的水平升高,谷树叶水提醇沉组的作用最为显著。谷树叶不同提取物组均可以不同程度地下调Bax蛋白表达、上调Bcl-2蛋白表达。结论谷树叶提取物能够降低高脂血症大鼠的血脂,并可以通过调节凋亡蛋白Bax和Bcl-2的表达来对抗高脂模型大鼠所致的血管内皮细胞凋亡。
        Objective To study the effect of extracts from Broussonetia papyrifera( L.) leaves on blood lipid of rats with hyperlipidemia and the expression of Bax,Bcl-2 in vascular endothelial cells. Methods 60 male SD rats were randomly divided into normal control group,model control group,fenofibrate group and extracts groups( water extraction,water extraction and alcohol precipitation,alcohol extraction),with 10 rats in each group. The models were established by high fat emulsion intragastrically and intervened with corresponding medicine for8 weeks. The levels of blood lipid were detected at 4 thand 8 thweek. The expression of Bax and Bcl-2 in vascular endothelial cells were detected. Results Compared with model group,TC and LDL-C decreased in extracts groups,and the level of HDL-C increased. The group of water extraction and alcohol precipitation has the most significant effect. The different extracts of Broussonetia folium groups had different degrees of down regulation of Bax protein expression and up regulation of Bcl-2 protein expression. Conclusion The extracts from Broussonetia papyrifera( L.) leaves can reduce the blood lipid level of hyperlipidemia and reduce the apoptosis of vascular endothelial cells in rats with hyperlipidemia through regulation of Bax and Bcl-2 expression.
