补气通络解毒方对胰腺癌模型小鼠肿瘤高甲基化基因1、沉默信息调节因子、Shh蛋白浓度的影响

摘要
目的观察补气通络解毒方对胰腺癌模型小鼠肿瘤高甲基化基因1(HIC1)、沉默信息调节因子(SIRT_1)、Shh蛋白浓度的影响。方法将30只裸鼠随机分为3组各10只,采用癌细胞种移植法造模。造模成功后,空白组和模型组给予生理盐水灌胃,治疗组给予补气通络解毒方灌胃,连续14 d。治疗结束后,处死裸鼠,取出瘤组织,以WB法检测各组标本的HIC1、SIRT_1、Shh蛋白浓度。结果 3组HIC 1、SIRT 1、Shh蛋白浓度比较有显著差异(P<0.05)。结论补气通络解毒方对模型小鼠HIC1、SIRT_1、Shh蛋白浓度有调节的作用。
        Objective To observe the effect of Buqi Tongluo Jiedu decoction on protein concentrations of HIC1、SIRT_1、Shh in model mice with pancreatic cancer. Methods Totally 30 mice were randomly and averagely divided into three groups. Mice with cancer were modeling by cancer cells seeding method. After successfulmodeling,the blank group and model group were given saline for gastric lavage,while the treatment group was given Buqi Tongluo Jiedu decoction for gastric lavage. All the three groups were treated for 14 days. After treatment,the mice were sacrificed for picking up the tumor tissue. Protein concentrations of HIC1、SIRT_1、Shh were detected by WB method.Results There were significant differences in the concentrations of HIC1,SIRT_1 and Shh among the three groups( P < 0. 05). Conclusion Buqi Tongluo Jiedu decoction play an important role in regulating protein concentrations of HIC1、SIRT_1、Shh in model mice with pancreatic cancer.
