摘要
为研究竹叶青蛇在中国台湾岛的扩散趋势,从Gen Bank上获取竹叶青蛇部分cytb基因序列,长度约655 bp,通过Seqman对序列进行格式转换、剪切与校正,再用MEGA6.0进行序列比对,统计碱基含量和变异位点。从内类群个体共定义了28个单倍型,其中1个来自福建武夷山,27个来自台湾岛。运用Microsoft Visio 2007软件绘制cytb部分序列统计简约网络图。结合生物地理学信息,发现早期竹叶青蛇基本是由北向南的扩散路线,可以初步推断在中国台湾地区竹叶青蛇大体是由北部(台北)向中南部山区扩散的总体趋势。用分子方法推断物种扩散,为新兴的分子生态学研究方法,可以广泛应用于未来系统发育地理学研究。
        The study aimed at investigating the spread directions of Trimeresurus stejnegeri in Taiwan island.655 bp cytb gene sequences of were obtained from Gen Bank.Seqman software and was used for the format conversion,shearing and correction of sequences,then the contents of bases and mutation loci were compared and counted by MEGA 6.0 software.The total 28 individual haploid types were defined in the species groups of which 1 from Fujian Wuyi mountain and the other 27 from Taiwan island.The statistical simple network diagram of partial cytb sequence was drawn by Microsoft Visio 2007 software.Combining with biological geography information,the results showed that the early route of the snakes was spread from north to south.Thus,the author preliminarily inferred that T.stejnegeri spread roughly from northern(Taipei)to central and southern mountains for the overall trend.The species diffusion was inferred by using molecular methods as an emerging molecular ecology research way,which could be widely used in the phylogeography research in the future.
