摘要
目的通过观察三黄凝胶对耳郭痤疮模型大鼠血清血管内皮生长因子(VEGF)、血小板源性生长因子(PDGF)含量的影响,探讨三黄凝胶预防痤疮瘢痕形成的机制。方法将36只6~8周龄Wistar大鼠随机分为空白组,模型组,阳性对照组,三黄凝胶低、中、高剂量组6组,每组6只,除空白组外,其余各组在大鼠右侧耳郭导管开口处均匀涂抹油酸0.5~1.0 m L/次,每日1次,隔日于大鼠耳郭皮内注射痤疮丙酸杆菌菌液,连续抹药3周,肉眼见囊肿即可停止造模,随机抽取2只模型组大鼠,分别剪下鼠耳,HE染色,进行病理学观察。实验第22天,阳性对照组以5 g/kg剂量外涂维A酸乳膏;三黄凝胶高、中、低剂量组分别以0.08,0.04,0.02 g/kg剂量外涂三黄凝胶;空白组和模型组外涂三黄凝胶基质。给药部位均为大鼠右耳郭造模处,每日1次,连续给药21 d。分别于造模第1,2,3周及用药第1,3周观察大鼠耳郭皮损变化;末次用药24 h后,大鼠心脏取血,离心,取血清置于-80℃冰箱中保存,采用ELISA法检测血清VEGF、PDGF的含量。结果与空白组比较,模型组血清VEGF、PDGF含量均明显升高,差异有统计学意义(P<0.01);与模型组比较,阳性对照组、三黄凝胶中剂量组及高剂量组血清VEGF、PDGF含量均明显下降,差异有统计学意义(P<0.05或P<0.01);与三黄凝胶低剂量组比较,三黄凝胶中剂量组血清VEGF、PDGF含量均明显下降,差异有统计学意义(P<0.05)。结论三黄凝胶预防痤疮瘢痕形成的机制可能与抑制和减少VEGF、PDGF的产生有关。
        Objective To observe the effects of Sanhuang Ningjiao( 三黄凝胶) on the serum levels vascular endothelial growth factor( VEGF) and platelet-derived growth factor( PDGF) levels in rats model with auricular acne,and to explore the mechanism of Sanhuang Ningjiao in preventing acne cicatrization.Methods Thirty-six 6 to8 week old Wistar rats were randomly divided into 6 groups,including blank group,model group,positive control group and the low,medium and high dose Sanhuang Ningjiao groups,with 6 rats in each group. Except the blank group,the rats in the other groups were applied evenly with oleic acid in doses of 0.5 to 1.0 m L once a day at the opening of right lateral auricular ducts,and propionibacterium acne solution was injected into rat auricular intracutaneously every other day,continuous application for 3 weeks. No sooner had been the cysts visible to the naked eyes than we stopped making models. Two rats in the model group were randomly chosen and their ears were cut off and stained with HE for pathological observation.On the 22 ndday of the experiment,the positive control group was externally applied with tretinoin cream in doses of 5 g per kg. The high,medium and low dose Sanhuang Ningjiao groups were externally applied with Sanhuang Ningjiao in doses of 0.08 g per kg,0.04 g per kg and 0.02 g per kg,respectively. The blank group and model group were applied with Sanhuang Niangjiao matrix.The application sites were at the right auricle of rats,once a day,successive medication for 21 days. The changes of auricle lesions were observed at the 1 st,2 ndand 3 rdweek of model establishment and at the 1 stand 3 rdweek of application respectively. After the last 24 hours of application,the blood was taken from the rat hearts,centrifuged,and the serum was stored in a refrigerator at-80 ℃. The levels of serum VEGF and PDGF were detected by ELISA.Results Compared with those of the blank group,the levels of serum VEGF and PDGF in the model group all increased significantly,and the difference was statistically significant( P<0.01); compared with those of the model group,the levels of serum VEGF and PDGF in the positive control group,the medium and high dose Sanhuang Ningjiao groups all decreased significantly,and the difference was statistically significant( P<0.05 or P<0.01). Compared with the low dose Sanhuang Ningjiao group,the levels of serum VEGF and PDGF in the medium dose Sanhuang Ningjiao group all decreased significantly,and the difference was statistically significant( P < 0. 05). Conclusion The mechanism of Sanhuang Ningjiao prevention on acne cicatrization may be related to inhibiting and reducing the production of VEGF and PDGF.
