摘要
目的:从车前草中提取分离车前草多糖,通过柱前衍生化高效液相法(HPLC)对车前草多糖的单糖和糖醛酸组成进行研究,建立车前草多糖特征图谱。方法:采用水提醇沉法分离,纯化车前草多糖,通过对车前草多糖PMP柱前衍生化,利用高效液相色谱法对多糖组分进行分析。结果:车前草多糖是由葡萄糖、甘露糖、鼠李糖、半乳糖、葡萄糖醛酸、半乳糖醛酸、木糖、阿拉伯糖等8种单糖组成,摩尔比为2.39∶0.20∶0.65∶2.05∶0.19∶0.91∶0.21∶1.08。方法学验证结果表明本实验测定方法精密度良好,样品回收率在97.906~98.658,相对标准差在1.17%~1.72%,单糖标准曲线线性相关系数均大于0.999,说明该实验建立的方法有效可信。结论:实验建立车前草多糖的分离提取方法和利用高效液相色谱法建立多糖特征图谱的研究方法,操作简便、分离度高,稳定性和重现性良好,该方法可用于车前草多糖的单糖组分分析和为其质量控制提供参考。
        Objective:The polysaccharide of psyllium was extracted from psyllium,and the polysaccharide composition of psyllium was studied by columnar derivatization and high performance liquid chromatography(HPLC). Methods:The polysaccharides were extracted by water method,and the polysaccharides were extracted by PMP column. The polysaccharide composition was analyzed by high performance liquid chromatography(HPLC). Results:The polysaccharide is composed of glucose,mannose,rhamnose,galactose,glucuronic acid,galacturonic acid,xylose and arabinose. The molar ratio was 2.39∶0.20∶0.65∶2.05∶0.19∶0.91∶0.21∶1.08. The results showed that the precision of the method was good,the recovery rate was between 97.906 and 98.658,the relative standard deviation was between 1.17% and 1.72%,and the linear correlation coefficient of the standard curve of monosaccharides was more than 0.999,which indicated that the experiment method was effective and credible. Conclusion:In this study,the method of separation and extraction of polysaccharide from psyllium and the method of monosaccharide composition analysis were established. The method was simple,stable,accurate and feasible. The method was applied to the analysis and determination of the content of polysaccharide in psyllium.
