桔梗、白花蛇舌草不同组分对人肺癌细胞A549的抑制作用

摘要
为筛选桔梗和白花蛇舌草中的抗肿瘤活性成分,制备了桔梗总黄酮、总皂苷提取物,白花蛇舌草总黄酮、总蒽醌提取物,采用亚硝酸钠-硝酸铝比色法检测总黄酮含量,香草醛法检测总皂苷含量,紫外吸收法检测总蒽醌含量,采用四甲基偶氮唑蓝比色法(MTT)检测4种提取物对人肺癌细胞A549的抑制作用。结果表明:桔梗和白花蛇舌草总黄酮含量分别为5.51%和1.19%,桔梗总皂苷含量为1.71%,白花蛇舌草总蒽醌含量为7.81%。桔梗总皂苷浓度为0.04 mg/mL时,对人肺癌细胞A549抑制率达到87.59%,桔梗总黄酮浓度为0.5 mg/mL时,抑制率达到89.16%。白花蛇舌草总蒽醌浓度为1.2 mg/mL时,抑制率达到34.76%;白花蛇舌草总黄酮浓度为0.3 mg/mL时,抑制率达到24.33%。试验结果显示,桔梗总黄酮和总皂苷对肺癌细胞A549的抑制作用较强,可为抗肿瘤药物的研究提供参考。
        To screen the antitumor active component of Platycodon grandiflorum(Jacq.) A.DC. and Hedyotis diffusa Willd, total flavonoids and total saponins of Platycodon grandiflorum, total flavonoids and totalanthraquinones of Hedyotis diffusa Willd were prepared. The content of total flavonoids was determined bysodium nitrite-aluminum nitrate colorimetric method, the content of total saponins was determined by vanillinassay, and the content of total anthraquinones was determined by ultraviolet spectromter method. Theinhibitory effects of 4 extracts on human lung cancer cells A549 were determined by methyl thiazolyltetrazolium(MTT) assay. The results showed that, the contents of total flavonoids from Platycodon grandiflorum and Hedyotis diffusa Willd were 5.51% and 1.19%, the content of total saponins from Platycodon grandiflorum was 1.71%, the content of total anthraquinones from Hedyotis diffusa Willd was 7.81%. The inhibition rate of0.04 mg/mL total saponins from Platycodon grandiflorum on human lung cancer cells A549 was 87.59%, theinhibition rate of 0.5 mg/mL total flavonoids from Platycodon grandiflorum was 89.16%, the inhibition rate of1.2 mg/mL total anthraquinones from Hedyotis diffusa Willd was 34.76%, the inhibition rate of 0.3 mg/mL totalflavonoids from Hedyotis diffusa Willd was 24.33%. Taken together, total flavonoids and saponins from Platycodon grandiflorum have strong antitumor activity, which provides some powerful reference for thedevelopment of related antitumor products.
