摘要
目的:探讨因子分析与聚类分析在综合评价补中益气汤(Buzhong Yiqi Decoction,BYD)对气虚发热大鼠免疫功能影响中的应用。方法:大鼠随机分成正常组、气虚发热组、阳性对照组(阿司匹林溶液0. 1 g·kg-1)、BYD高剂量组(25. 34 g·kg-1)、BYD中剂量组(12. 67 g·kg-1)、BYD低剂量组(6. 34 g·kg-1),在限食法的基础上采用番泻叶(0. 1 g·kg-1)苦寒泻下加游泳劳倦过度及注射脂多糖(LPS,80μg·kg-1)的方法建立气虚发热大鼠模型。造模42 d后连续给药6 d,末次给药后注射LPS,检测与免疫功能相关指标,包括CD4+、CD8+细胞百分比、补体C3水平、免疫球蛋白Ig M、Ig G水平及淋巴细胞转化率,并运用因子分析与聚类分析综合评价BYD对气虚发热大鼠免疫功能的影响。结果:BYD能显著改善气虚发热大鼠的免疫功能,其中低剂量效果最显著,中剂量与高剂量次之,阿司匹林效果较差。结论:因子分析与聚类分析对气虚发热大鼠的免疫功能进行综合评价是可行的,可作为评价中药复方疗效科学、客观的新方法。
        OBJECTIVE To explore the application of factor analysis and cluster analysis in the comprehensive evaluation ofthe effect of Buzhong Yiqi Decoction( BYD) on immune function in rats with deficiency of vital energy and fever. METHODSRats were randomly divided into normal group,deficiency of vital energy and fever group,positive control group( aspirin solution0. 1 g·kg-1),BYD high dose group( 25. 34 g·kg-1),BYD medium dose group( 12. 67 g·kg-1),BYD low dose group(6. 34 g·kg-1). Deficiency of vital energy and fever model was established by combination of limiting diet,folium sennae bittercold diarrhea( 0. 1 g·kg-1),swimming over fatigue and injection of lipopolysaccharide( LPS,80 μg·kg-1) in rats. After themodel was established for 42 days,corresponding drug was administered continuously for 6 days. LPS was injected after the lastadministration,and then the immune function related indexes were detected,including the percentage of CD4+,CD8+cells,complement C3,immunoglobulin IgM,IgG level,and lymphocyte transformation rate. Finally,the effect of BYD on immunefunction in rats with deficiency of vital energy and fever was comprehensively evaluated by factor analysis and cluster analysis.RESULTS BYD significantly improved the immune function of the rats with deficiency of vital energy and fever,of which thelow dose effect was the most significant,the middle dose and the high dose were the next,and aspirin was relatively ineffective.CONCLUSION Factor analysis and cluster analysis are feasible for comprehensive evaluation of immune function in rats withdeficiency of vital energy and fever. It can be used as a scientific and objective method to evaluate the efficacy of traditional Chi-nese medicine compound prescriptions.
