摘要
目的以高效液相色谱法测定番石榴、番荔枝、大红柿中的没食子酸、儿茶素、表儿茶素、绿原酸、咖啡酸、阿魏酸、槲皮素、木犀草素、山奈酚共9种多酚类物质。方法用Diamosil(R)-C18(250 mm×4.6 mm,5μm)为分离柱,以甲醇和0.02%(V/V)甲酸水溶液(p H=3.0)为流动相进行梯度洗脱,流速为0.8 ml/min,进样量为10μl,柱温为35℃,二极管阵列检测器检测。结果 9种多酚类物质在40 min内完全分离,方法在0.25 mg/L~50 mg/L时线性良好,相关系数为0.999 8~0.999 9;检出限(S/N=3)为0.029 mg/L~0.18 mg/L;样品回收率为93.6%~101%,相对标准偏差为1.34%~6.99%。结论番石榴和番荔枝中检出酚类物质各6种,大红柿中检出酚类物质5种。该测定方法准确、灵敏,适用于番石榴、番荔枝、大红柿中9种多酚物质含量的检测。
        Objective High performance liquid chromatography( HPLC) method was adopted to determine 9 types of polyphenols,including Gallic acid,( +)- Catechin,(-)- Epicatechin,Chlorogenic acid,Caffeic acid,Ferulic acid,Quercetin,Luteolin and Kaempferol in guava,cherimoya,and persimmon. Methods Diamosil( R)- C18( 250 mm × 4. 6 mm,5 μm)was adopted as the separation column; methanol and 0. 02%( V / V) formic acid solution( p H = 3. 0) were used as mobile phase for gradient elution. The flow rate was 0. 8 ml / min,the injection volume was 10 μl and the column temperature was 35 ℃.Diode array detector was used for detection. Results 9 types of polyphenols were separated completely within 40 min. It showed a good linearity when the method was performed within 0. 25 mg / L- 50 mg / L,and the correlation coefficient( r) was within 0. 999 8- 0. 999 9. The detection limit( S / N = 3) was within 0. 029 mg / L- 0. 18 mg / L. The recovery rate was within93. 6%- 101%,and the relative standard deviation( RSD) was within 1. 34%- 6. 99%. Conclusion 6 types of polyphenols were detected from guava and cherimoya,and 5 types of polyphenols were detected from persimmon. The determination method above is accurate and sensitive,and it is a suitable method to determine 9 types of polyphenols in guava,cherimoya and persimmon.
